目的:分析、比较姜黄、郁金和莪术等3种姜黄属中药中多种活性成分对人脐静脉内皮细胞(human umbilical vein endothelial cell,HUVEC)生长、增殖的抑制作用,以期筛选出活性最强的抗血管生成成分.方法:①采用噻唑兰(3-(4,5-dimethylthiaaol-2-y1)-2,5-diphenyl tetrazoli-am bromide,MTT)还原实验比较3种姜黄属中药多种活性成分对人脐静脉内皮细胞生长、增殖的抑制作用.②采用流式细胞术(flow cytome-try,FCM)实验探讨活性成分对内皮细胞及小细胞肺癌细胞株SPC细胞增殖周期影响的差异性.结果:①3种姜黄属中药挥发油均可抑制人脐静脉内皮细胞的增殖;姜黄挥发油的生物学活性明显高于郁金和莪术,差异有显著性意义(x2=6,P=0.014).②3种醇提物中仅姜黄醇提物对内皮细胞的生长有显著抑制作用.③3种姜黄素单体抑制内皮细胞增殖的生物活性具有显著差异:姜黄素-3＞姜黄素-2＞姜黄素-1,差异有显著性意义(x2=5～7,P=0.008～0.025).④浓度为4 mg/L时姜黄素-3可将内皮细胞阻滞于S期;姜黄素-3的浓度增至8mg/L以上时不但引起S期细胞积聚更加显著,而且出现明显的诱导凋亡作用.但浓度为10 mg/L的姜黄素-3对人小细胞肺癌细胞系SPC的增殖周期仍无明显影响.结论:姜黄色素和姜黄挥发油可能是姜黄属中药抗血管生成作用的主要活性成分;姜黄素-3对内皮细胞增殖周期的抑制作用具有细胞种类特异性.
AIM:To analyze and compare the inhibitive effects of fragments from three spec ies of plant belonging turmeric genus(curcuma langa,aromatic turmeric root-tube r and zedoary) on the growth and proliferation of human umbilical vein endotheli al cells(HUVEC),so as to screen out the most active fragment against the vascula rization. METHODS:①3-(45-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide m ononuclear cell direct cytotoxicity assay(MTT) was used to analyze the inhibitiv e effect of active fragments from three species of plant belonging to turmeric g enus on the growth and proliferation of HUVEC.② Flow cytometry(FCM) was used t o investigate the effect of three monomes of curcumins from curcuma langa on the cell cycle of HUVEC. RESULTS:①The volatile oils from the three species of plant belonging to turme ric genus could inhibit the proliferation of HUVEC,the biological activities of volatile oils of curcuma langa were obviously higher than those of aromatic turm eric root-tuber and zedoary(χ2=6,P=0.014).②Among three species of extract, on ly the extract from the main root of curcuma langa had remarkably inhibitive eff ect on the growth of HUVEC.③At the concentration of 4 mg/L,there were significa nt differences in the biological activities of the three species of plants belon ging to turmeric genus for inhibiting the proliferation of HUVEC:curcumin-3 >cu rcumin-2 >curcumin-1(χ2=5-7,P=0.008-0.025).④At the concentration of 4 mg/L ,curcumin-3 could effectively retard the endothelial cell at the S phase.And wh en the concentration was increased to 8 mg/L or more, curcumin-3 could also ind uce obvious apoptosis of HUVEC;However,at the concentration of 10 mg/L,curcumin -3 had no effect on the proliferative cycle of human small cell lung cancer cel l line SPC. CONCLUSION:Both of volatile oils from the three species of plants belonging to turmeric genus and the segments of curcuma langa may be the main fragments of a nti-vascularization of the herbs belonging to turmeric genus.The inhibitiv
