Objective:To establish a new animal model with cerebral palsy and identify the animal model.Method: 24 Wistar pregnant rats were divided into two groups: lipopolysaccharide(LPS) (n=16) and normal saline(n=8). Pregnant rats were consecutively injected intraperitoneally with LPS (350μg/kg) or saline on gestation 16d and 17d. The interval from infusion to delivery for each group was recorded, as well as their birth weight. The abilities of sensorimotor movement (hanging test and inclined plane test),emotional behavior(open field test and resistant to capture test), posture, and muscle tension, et al were tested till 30d. The brain damage of the preterm rats was observed by optical and electron microscopes.Result:There were 26 live preterm pups delivered by 6 LPS-treated rats.The birth weight of the pups reduced compared with the saline controls.And there was motor reduction of preterm pups.There was intrauterine infection of LPS group rats.There was white matter damage of animal modal. 10 of 16 preterm pups were the ones with cerebral palsy.The behavioral abilities of CP pups were much lower than those of normal control group.There was much abnormality of posture and muscle tension with CP pups.Conclusion: Preterm delivery can be induced by LPS.A new animal model for investigation of cerebral palsy is established by LPS-induced preterm.A method for identifying rats with cerebral palsy is successfully established. The damage location of the CP animal model is white matter.
