Diethylaminoethyl hydroethylate (DEAE-E/H), a kind of weakly basic macroporous anion resin, was used as the carrier to immobilize the aminoacylase from Aspergillus oryzae by ion-combination method. The specific activity of the immobilized aminoacylase on DEAE-E/H (IME) was up to 1350U/g. The properties of the immobilized aminoacylase on DEAE-E/H (IME) were investigated. The results showed that the IME had high thermostability. Compared with free aminoacylase, the appropriate reaction conditions of IME was changed, the reaction temperature changed from 47°C to 57°C, pH changed from 7.0 to 6.5, and the proper reaction condition range was increased. It was also proved that Co2+ was a good activator for IME at the concentration of 1 × 10-3 mol/L. Additionally, the stabilities of IME in batch and continuous operation were studied too. The activity loss was only 5% after 3 times used in batch reactor and the half-time of IME in continuous operation was up to 20 days. It illustrated that the IME had good operational stability.
