亚麻转基因中抗生素应用效果的研究 预览 被引量:12

摘要 在利用农杆菌介导法进行亚麻抗除草剂草丁膦(Basta)目的基因(Bar)导入工作中;对选择培养基内加入500mg/l克拉维酸钾(timentin进口)+200mg/l头孢噻肟(claforan进口)的抗菌素来取代500mg/l头孢噻肟(Cefotaxime国产)不仅脱菌效果彻底,期效长,且不影响亚麻愈伤形成和芽的分化;同时对再生植株进行了盆栽Basta抗性筛选试验,获得不同抗性的植株和种子. The antibiotic effects of Timentin,Claforan and cefotaxime were tested in the transformation of bar gene which is resistance to herbicide Basta by Agrobacterium tumefaciens . Compared with 500mg/l cefotaxime,the effect of 500mg/l timentin+200mg/l is better in the selective medium.It not only controlls bacteria completely and keeps long time,but also does't influence the forming of calli and the growing of buds. At the same time,the effect of resistance to Basta was tested, the regeneration plants and seeds of resistant different concentration of Basta have been obtained.
