泽兰化学成分反相高效液相色谱分离条件的优化 预览 被引量:2

摘要 优化了泽兰V(乙醇):V(水)=70:30提取物的分离条件。采用反相高效液相色谱法(RP—HPLC)考察了不同波长、酸抑制剂类型及其浓度、流动相梯度、进样量以及柱温对其分离效果的影响。确定其分离条件如下:Hypersil ODS2色谱卡柱(5μm,4.6mmi.d.×250mm);流速1.0mL/min;检测波长280nm;流动相:(A)击(甲酸)=0.75%的水溶液(以下简称为X%Y水,其中X为数字,Y为水中的添加剂),(B)乙腈;梯度洗脱条件为:φ(B)=10%(5min)→^65min φ(B)=55%;进样量为20μL;柱温30℃。该方法获得的色谱图基线校平稳,分离度较高,能有效地对泽兰化学成分进行分析,可用于泽兰的质量控制。 The separation conditions for the extract of Herba Lycopi was optimized using reverse phasehigh performance liquid chromatography ( RP - HPLC ). The separation was. performed by a Hypersil ODS2 column(5 μm,4.6 mm i. d. ×250 mm)at a flow rate of 1.0 mL/min with the detection wavelength at UV 280 nm. The mobile phase consisted of water with formic acid( 100:0. 75) (A) and acetonitrile (B). The gradient profile was that solution Bwas held 10% for 5 min and then linearly from 10% -55% in 65 min. The injection volume was 20 μL and the column temperature was maintained at 30 ℃. The chromatogram obtained with the above method was provided with smoother baseline and higher separation resolution. The method could be well used for the component analysis and the quality control of Herba Lycopi.
