目的研究恶性肿瘤患者病理组织中人纤维介素（human fibrinogen-like protein2，hfgl2）的表达及与纤维蛋白沉积的关系。方法采用免疫组化方法检测了11例结肠癌、15例宫颈癌、10例乳腺癌、9例食管癌、12例肺癌、16例胃癌患者的病理组织切片中hfgl2及纤维蛋白的表达，分析二者的组织学关系，用RT-PCR方法检测了部分肿瘤组织与肿瘤细胞系中的hfgl2 mRNA。结果免疫组化显示hfgl2在上述肿瘤中均有表达，主要分布于间质浸润细胞、肿瘤实质细胞、肿瘤血管内皮细胞及细胞外基质，且在邻近有明显的纤维蛋白沉积。通过RTPCR，在体外培养的数种肿瘤细胞系中未检出hfgl2 mRNA，但在肿瘤组织中检出hfgl2 mRNA表达明显高于正常组织。结论恶性肿瘤时存在hfgl2的高表达，并且可能与肿瘤高凝状态的形成及肿瘤的演进有关。
Objective To investigate the protein expression of human fibroleukin （hfgl 2） and focal fibrin deposition in cancer and its potential clinical significance. Methods Malignant tumor tissues were obtained from 11 patients with colon cancer, 15 patients with cervical cancer, 10 patients with breast cancer, 9 patients with esophageal cancer, 12 patients with lung cancer and 16 patients with gastric carcinoma. Nonmalignant tissue was used as negative control. Three tumor cell lines were also studied. Hfgl 2 was examined both at the level of mRNA and protein by immunohistoehemistry as well as RT-PCR. Double staining was also used in the hfgl 2 positive samples to examine the association of hfgl 2 and fibrin expression. Results Immunohistochemistry showed that hfgl 2 was expressed in 67 out of 73 patients from 6 different kinds of cancers, mainly distributed in tumor parenchyma cells, interstitial infiltrated cells and vascular endothelial cells. The hfgl 2 expression was closely associated with local fibrin deposition in 6 kinds of studied cancers. RT-PCR revealed that hfgl 2 mRNA was up-regulated in tumor tissues as compared with that in controls. Hfgl 2 was not evidenced in 3 tumor cell lines cultured in vitro. Conclusion The expression of hfgl 2 may be correlated with the characteristic hypercoagulability in cancer, which in turn induces tumor angiogenesis and metastasis. The molecule of hfgl 2 may serve as a potential therapeutic target for the management of tumor development.
