HPLC法测定黄芩提取物中4种主要活性成分的含量 预览 被引量:8

Determination Four Active Ingredients of Extraction from Scutellaria Baicalensis Georgi by HPLC

摘要 以建立黄芩提取物中黄芩苷、汉黄芩苷、黄芩素和汉黄芩素含量的高效液相色谱(HPLC)测定方法为目的,通过下列方法:色谱柱为Spherigel—C18柱(5μm,4.6mm×200mm),流动相:0.2%醋酸(A)-甲醇(B),梯度洗脱:0~30min,B:42%-70%;流速1.0ml/min;检测波长276nm,测得黄芩苷、汉黄芩苷、黄芩素、汉黄芩素的线性范围分别为:0.500~2.00μg(r=0.9996)、0.66-3.30μg(r=0.9995)、0.55—2.75μg(r=0.9998)、0.51~2.55μg(r=0.9994);黄芩苷、汉黄芩苷、黄芩素和汉黄芩素平均回收率(n=5)分别为:98.32%、99.17%、101.25和102.53,RSD分别为2.65%、2.48%、1.92%和3.04%(n=5)。该方法简便、准确,可用于黄芩提取物的质量控制。 To establish a HPLC method for determination of baicalin,baicalein,wogonoside and wogonin in extraction Scutellaria baicalensis Georgi. The analytical column was Spherigel-C18 column(5 μm, 4.6×200 mm ). The mobile phases was 0.2 % HAc(A)-methanol(B) at a flow rate 1.0 ml/min. Gradient elution, 0-30min: B 42%-70%. The detection wavelength was 276 nm. The linear ranges of baicalin, wogonoside, haicalein and wogonin were 0.500-2.00 μg(r =0.9 996), 0.66-3.30 μg (r =0.9 995),0.55-2.75 μg (r =0.9 998),0.51-2.55 μg (r =0.9 994).The average recoveries (n=5) of baicalin,wogonoslde, baicalein and wogonin were 98.32%, 99.17%, 101.25, 102.53, respectively; RSD were 2.65%, 2.48%, 1.92% , 3.04%, respectively. The method is simple,accurate and can be used to determine active ingredients of extraction from Scutellaria baicalensis Georgi.
