Objective： To investigate CYP19 mRNA and protein expression in ovary tissue of rat model of letrozole-induced polycystic ovary syndrome （PCOS）, and to provide experimental data for the model application. Methods： Forty rats were divided into two groups randomly, PCOS group administered letrozole once daily during 21 and a control group. Gonadal hormone concentrations in serum were determined by radioimmunoassay, the histologie changes in ovaries were observed by H-E staining, and the expression of CYP19 gene in ovary tissue was detected by Real time-PCR, Western blot and immunohistochemistry. Results： Compared to the control group, estradiol and progesterone showed a considerable reduction （P〈 0. 05）. Levels of serum testosterone, FSH and LH in the model group were markedly increased than that in the control group （P%0.05）. The expressions of CYP19 mRNA and protein were significantly up-regulated in the model group than those in the control group. Conclusion： The expression of CYP19 gene in letrozole-induced polyeystic ovary is significantly higher than that in the control ovary.
