p53在异丙肾上腺素所致心肌纤维化大鼠心肌组织中的表达及意义 预览

摘要 目的:探讨p53在异丙肾上腺素(Iso)所致心肌纤维化大鼠心肌组织中的表达及意义,阐明其在心肌纤维化中的作用。方法:25只大鼠随机分为5组,正常对照组及注射Iso后12h、24h、1周和3周组。除正常对照组外,其他各组皮下多点注射Iso(15mg·kg-1)复制大鼠心肌损伤模型。采用免疫组织化学染色及RT-PCR分别检测心肌p53蛋白及基因表达。结果:免疫组织化学染色显示,注射Iso后12和24h心肌P53蛋白表达水平明显高于正常对照组(P〈0.05),注射1和3周后p53蛋白表达增加更明显(P〈0.01)。RT-PCR结果显示,与正常对照组比较,注射Iso后12h心肌组织中p53mRNA表达明显增高(P〈0.05),注射Iso后24hp53mRNA表达水平达到最高(P〈0.01),而注射Iso后1周心肌p53mRNA表达较注射后24h有所下降,但仍高于正常对照组(P〈0.05)。结论:P53蛋白在大鼠损伤心肌中呈持续高表达,p53基因表达上调,提示p53在心肌纤维化形成过程中起一定作用。 Objective To investigate the expression of p53in myocardium tissue of ispproterenol(Iso)-induced myocardial fibrosis rats and its signigicance and clarify the effect in inyocardial of fibrosis.Methods 25male Wistar rats were randomly divided into five groups:control group,model groups(12h,24h,1week and 3weeks)(n= 5).Except control group,all the rats in the other groups were treated with Iso to bulit rat myocardial injury models (15mg·kg-1).The expressions of P53protein and p53 mRNA were detected with immunohistochemical staining and RT-PCR.Results The immunohistochemical results showed that the P53protein expressions were increased 12and 24hafter Iso injection compared with control group (P0.05)and the protein expressions were obviously increased 1week and 3weeks after Iso injection (P0.01).The RT-PCR results showed that the p53 mRNA was obviously increased at 12h(P0.05),arrived a peak at 24h(P0.01)and was gradually decreased at 1week, but still higher than normal level(P0.05).Conclusion The P53protein expression is increased in rat myocardium tissue and the expression of p53gene is up-regulated,suggesting that p53has an important role in rat myocardial fibrosis.
