几种鲜切花RNA提取方法的比较 预览 被引量:3

Extraction methods of total RNA from some cut flowers

摘要 以5种鲜切花为材料,分别采用Trizol法、异硫氰酸胍法、改良十六烷基三甲基溴化铵(Cetyltrimethyl ammonium bromide,CTAB)法和RNA plant试剂盒法提取总RNA,并运用琼脂糖凝胶电泳和紫外分光光度计检测比较RNA的完整性和纯度.结果表明,Trizol法和异硫氰酸胍法对百合(Lilium longiflorum)、非洲菊(Gerbera jamesonii)和香石竹(Dianthus caryophyllus)鲜切花RNA的提取效果较好,但不适于月季(Rosa hybrid)和姜花(Hedychium coronarium)的RNA提取.改良CTAB法和RNA plant试剂盒法对鲜切花RNA的提取具有普遍适用性,提取的RNA完整性好、纯度高、不易降解.其中改良CTAB法还具有成本低、提取量大等优点,是提取月季、百合、白姜花、非洲菊和香石竹鲜切花组织RNA的较理想方法. Four RNA isolation methods were compared concerning their adaptability to five cut flowers species(carnation,rose,lily,white ginger lily and gerbera) which were rich in polysaccharide and polyphenol.The results showed that Trizol and isothiocyanate methods were more suitable for RNA extracting from lily,gerbera and carnation cut flowers than from rose and white ginger lily.Improved Cetyl trimethyl ammonium bromide(CTAB) method and RNAplant reagent were found to be efficient for RNA extracting from all the five cut flowers.Additionally,improved CTAB method was more favorable for its cost efficiency and high yield.
