可转形穴位注射药线对脑缺血再灌注大鼠脑皮质MMP-9的影响 预览

The Effect of Deformable Embedding Medicine in Acupoints on Brain Tissue MMP-9 of Rats with Focal Cerebral Ischemia Reperfusion Injury

摘要 目的观察并比较可转形穴位注射药线(川芎嗪)和川芎嗪药物羊肠线对局灶性脑缺血再灌注大鼠脑皮质基质金属蛋白酶-9(MMP-9)表达的影响,以筛选新型的、便于临床使用的药线剂型。方法 SD大鼠随机分为模型组、川芎嗪药物羊肠线组和可转形穴位注射药线组,采用改良线栓法建立局灶型脑缺血(MCAO)再灌模型,在大椎、双侧内关穴埋药线,免疫组化法检测各组大鼠脑皮质MMP-9的表达。结果模型组大鼠脑皮质MMP-9表达升高,两治疗组大鼠皮质MMP-9表达均有降低,与模型组同时相比较有显著差异(均P〈0.05);可转形穴位注射药线组与川芎嗪药物羊肠线组同时相比较,1d组无明显差异(P〉0.05),3d组和5d组差异较显著(均P〈0.05)。结论穴位埋植可转形穴位注射药线可降低脑缺血再灌大鼠脑皮质MMP-9的水平,减少缺血再灌注损伤,其作用优于药物羊肠线,可作为新型的缓释药线应用于临床。 Objective The purpose of this research lies in selecting a new and convenient for clinical practice medicine,through comparing the effects of deformable embedding medicine in acupoints(collagen tetramethylpyrazine) and collagen tetramethylpyrazine catgut,and providing experimental evidence for clinical curing focal cerebral ischemia with deformable embedding medicine in acupoints.Methods 54 nos.rats are randomly divided into three groups which are model group,collagen tetramethylpyrazine..."catgut group and deformable embedding medicine in acupoints group.The focal CIR models were established by improved Zea-Longa and then embedding medicine in Dazhui and Neiguan acupoints with collagen tetramethylpyrazine sustained-release dose.The expression of MMP-9 in brain tissue is tested with the method of immunohistochemistry in groups.Results The expression of MMP-9 in brain tissue for model group increase only.But for the two curing groups,the expressions of MMP-9 in the brain tissue decrease.Compared with model group,expressions of MMP-9 in the brain tissue in other curing groups are fairly significant(P0.05).After three days and five days the comparison of collagen tetramethylpyrazine catgut group and deformable embedding medicine in acupoints group lead to a significant difference(P0.05).Conclution Embedding deformable medicine in acupoints can reduce the content of MMP-9 in the brain tissue of focal CIR models to minimize the damage to blood vessel during cerebral ischemia reperfusion.Its clinical function is superior to collagen tetramethylpyrazine catgut group.Deformable embedding medicine in acupoints(collagen tetramethylpyrazine)can be a new sustained-release embedding medicine in clinical practice.
