Research on Learning and Memory Ability of Vascular Dementia Rats Using Morris Water Maze

摘要 目的:采用Morris水迷宫实验比较不同造模方法制备的血管性痴呆大鼠的学习记忆能力。方法:采用单侧反复夹闭、单侧永久性结扎和双侧中一侧永久性结扎,另一侧反复夹闭大鼠颈总动脉3种方法造成大鼠血管性痴呆模型,采用Morris水迷宫图像自动采集和软件分析系统记录逃避潜伏期、平台穿越次数、各象限中游泳时间及距离。结果:空间定位航行试验中双侧造模大鼠的逃避潜伏期比正常假手术组大鼠的逃避潜伏期长,统计学比较有显著差异(P〈0.01),两组单侧造模大鼠的逃避潜伏期与正常假手术组大鼠的逃避潜伏期比较差异不明显。空间探索试验中双侧造模大鼠平台穿越次数比正常假手术组的平台穿越次数少,统计学比较有显著差异(P〈0.05),并且在第一、三象限中游泳距离均有不同差异。两组单侧造模大鼠的平台穿越数与正常假手术组比较无显著差异。结论:双侧造模方法更有利于研究血管性痴呆大鼠的空间学习记忆。 Objective:To compare the results on learning and memory ability of the vascular dementia(VD)in rats using Morris water maze.Methods:The model rats were made by three ways(clipping unilateral occlusion carotid arteries repeatedly,permanent ligation of unilateral common carotid arteries,permanent ligation of unilateral common carotid arteries and clipping the other side repeatedly).The Morris water maze tracking and analyzing system of the experimental animal trace was adopted to record the latency,times across the platform,swimming distance and time during platform quadrant. Results:Compared the latency in the test-looking for hidden platform,bilateral model rats had longer latency than that in sham -operated group,and there was significant difference between them(P0.01),there was no difference among the others.In spatial probe test,the times of passing through the platform belonged to bilateral model rats was less than that of sham-operated group,and there was significant difference(P0.05 ).The distance of swimming test was obviously different in the four quadrants.Conclusion:The method of bilateral model was more desirable in the research of VD rats in space learning and memory.
