醇化过程中烤烟烟叶表面可培养真菌的多样性 预览

摘要 为研究醇化过程中烤烟烟叶表面可培养真菌的多样性,选取国内外25个不同产地、等级的正常、霉变烟叶为研究对象,利用平板分离和分子生物学方法对烟叶表面的可培养真菌进行鉴定和系统发育分析。从415株真菌中挑选具有代表性的77株真菌进行ITS分析。结果表明:①从霉变烟叶中分离到的真菌,其多样性高于正常烟叶。②除7株真菌无法准确鉴定外,其余70株真菌分布在3个门、8个纲和30多个属中。优势菌属为曲霉属(Aspergillus)、分子孢子菌属(Cladosporium)和根霉属(Rhizopus),占比分别为22.9%、7.1%和7.1%。③从进口烟叶中分离到了特异的属,包括短梗霉属(Aureobasidium)、Myrmaecium和锥毛壳属(Coniochaeta)。表明醇化过程中烤烟烟叶表面的可培养真菌的多样性高,且真菌类别与烟叶产地密切相关。控制曲霉属真菌的生长繁殖是降低醇化烟叶霉变发生率的关键。 To investigate the diversity of culturable fungi on the surface of tobacco leaves during aging,25 domestic and imported_samples of normal and mildewed tobacco leaves of different qualities were collected. Plate isolation and molecular biology methods were adopted to identify and analyze the culturable fungi on tobacco leaves.In total of 415 culturable fungi were identified,and the ITS sequences of 77 representative strains were analyzed.The results showed that:1) The diversity of fungi on mildewed leaves was higher than that on normal leaves.2)Except for 7 uncertain strains,the other 70strains belonged to 3 phyla,8 classes and more than 30 genera.The dominant genera were Aspergillus,Cladosporium and Rhizopus,accounting for 22.9%, 7.1%and 7.1%,respectively.3) The unique genera (Aureobasidium,Myrmaecium and Coniochaeta)were isolated from imported tobacco leaves during aging.These results indicated that the diversity of culturable fungi on the surface of flue-cured tobacco leaves during aging was high and the category of fungi more closely correlated to tobacco growing areas.Furthermore,limiting the growth and reproduction of Aspergillus was a key step for reducing the incidence of mildew on tobacco leaves during aging.
