加味益气聪明汤对血管性痴呆大鼠行为学及皮层区单胺类神经递质的影响

The Influence of Flavored Qi-Boosting Intelligence Decoction on Ethology and Monoamine Neurotransmitters in the Cortex of Rats with Vascular Dementia

摘要 目的:通过建立血管性痴呆动物模型,探讨加味益气聪明汤对血管性痴呆(vascular dementia,VD)大鼠行为学及大脑皮层单胺类递质的影响。方法:将60只大鼠随机分为对照组15只和实验组44只,实验组采用双血管阻断法制备VD模型,对照组仅分离出颈总动脉但不结扎,随即缝合。将造模成功的44只大鼠随机分为模型组、西药组、中成药组、中药组,每组各11只。中药组予加味益气聪明汤14. 58 g·(kg·d)^-1灌服;西药组予以尼莫地平水溶液6. 25 g·(kg·d)^-1灌服,中成药组予以复方苁蓉益智胶囊水溶液0. 375 g·(kg·d)^-1灌服,模型组、对照组予以2 mL·d^-1蒸馏水灌服。选用Morris水迷宫实验检测大鼠的行为学变化,采用荧光光度法测定大脑皮层区多巴胺(dopamine,DA)、5-羟色胺(5-hydroxy tryptamin,5-HT)、去甲肾上腺素(norepinephrine,NE)的含量。结果:与对照组比较,模型组大鼠逃避潜伏期明显延长,差异具有统计学意义(P <0. 01),经过有效区及平台的次数显著减少,平台及有效区停留时间明显缩短(P <0. 01);西药组、中成药组以及中药组大鼠潜伏期有所延长,经过有效区及平台的次数减少,平台及有效区停留时间缩短,但差异无统计学意义(P>0. 05)。与对照组比较,模型组大鼠皮层区5-HT、DA、NE含量均明显降低,差异具有统计学意义(P <0. 01);西药组、中成药组及中药组较对照组VD大鼠皮层区5-HT、DA、NE显著降低,差异具有统计学意义(P<0. 01)。与模型组比较,西药组、中成药组及中药组VD大鼠皮层区5-HT、DA、NE含量均明显增高,差异具有统计学意义(P <0. 01)。结论:加味益气聪明汤能提高大脑皮层区NE、DA、5-HT的含量,调节传递效能,从而改善大鼠学习记忆能力。 Objective: To explore the influence of Flavored Qi-Boosting Intelligence Decoction on ethology and monoamine neurotransmitters in the cortex of rats with vascular dementia by establishing animal model of vascular dementia. Methods: Sixty rats were randomly divided into the control group with 15 rats and the experimental group with 44 rats. The VD models were made by double vessel occlusion in the experimental group. The common carotid artery was isolated but not ligated in the control group,and then sutured. Forty-four rats with successful modeling were randomly divided into the model group,the western medicine group,the Chinese patent medicine group and the TCM group,with 11 rats in each group. The TCM group was given 14. 58 g·( kg·d)-1 of Flavored Qi-Boosting Intelligence Decoction for oral administration. The western medicine group was given 6. 25 g·( kg·d)-1 of Nimodipine solution for oral administration. The TCM group was given compound 0. 375 g·( kg·d)-1 of Zhirong Yizhi Capsule solution,and the model group and the control group were given 2 m L·d-1 of steaming water for oral administration. Morris water maze test was used to detect the behavioral changes of rats. The contents of dopamine( DA),5-hydroxy tryptamin( 5-HT)and norepinephrine( NE) in cerebral cortex were determined by fluorescence spectrophotometry. Results: Compared with the control group,the escape latency( EL) of the model group was significantly prolonged,and the difference was statistically significant( P < 0. 01). The number of times passing through the effective area and platform was significantly reduced,and the residence time of the platform and effective area was significantly shortened( P < 0. 01). The EL of rats in western medicine group,the Chinese patent medicine group and the TCM group was prolonged,the number of effective areas and platforms was reduced,and the residence time of platforms and effective areas was shortened,but there was no significant difference( P > 0. 05).
