云南省2例人感染H5N6禽流感病毒的基因特征研究

Genetic characterization of avian influenza A(HSN6)virus from two patients in Yunnan province

摘要 目的分析云南省2例人感染H5N6禽流感病毒的基因特征,为将来的H5N6流感病毒的预防和控制提供科学依据。方法采集云南省2例肺炎患者的咽拭子以及居住地禽类相关外环境标本,进行禽流感病毒H5N6的基因检测,并将阳性标本进行高通量测序以获得全基因组序列。利用MEGA软件进行序列比对,分析H5N6禽流感病毒HA基因的功能性位点变异情况,并构建HA基因系统发育树进行分析。结果全基因组序列分析结果显示,云南省2例人感染H5N6禽病毒的8个基因片段序列同源性为98.5%~100.0%之间,与四川省、广东省的人感染H5N6禽流感病毒基因比对分析发现,除NP基因外,其他7个片段均有功能性位点的差异,导致其受体特异性改变、病毒毒力增强及耐药性改变。HA基因进化树分析显示,云南株、四川株及广州株同属于H5亚型2.3.4分支中的2.3.4.4亚支,云南省内外环境分离的6株禽类H5N6病毒与云南省2例人H5N6病毒及广州株有一定的亲缘关系,归于同一大簇。四川株与云南株及广州株有一定的遗传距离。结论云南省H5N6禽流感病毒在遗传中存在着多样性和复杂性,病毒基因功能性位点的变异可以是引起禽流感病毒跨种传播的机制之一。因此,应动态监测活禽市场中的禽流感病毒的分布情况及基因变异特征,为研究禽流感病毒跨种传播提供研究资料,同时为云南省禽流感病毒的防控提供科学依据。 ObjectiveTo analyze the genetic features for two human avian influenza A(H5N6) virus strains from patients in Yunnan province, and to provide the scientific basis for the prevent and control of avian influenza virus A(H5N6) in the future.MethodsThroat swab samples from two patients with pneumonia and samples from livestock near the patients' houses in Yunnan province were collected to detect avian influenza A(H5N6) virus. Whole genome sequences of the strains were obtained from the positive samples with high throughput sequencing. Sequence alignment with MEGA software was used to analyze mutations in functional loci, and the phylogenetic tree of hemaglutinin (HA) gene was constructed.ResultsWhole genome analysis showed that the similarities of the 8 RNA fragments in the two virus stains from Yunnan province were 98.5%-100.0%. Comparing to the strains from Sichuan and Guangzhou, differences in functional loci were found in all seven fragments of Yunnan strains except for NP fragment. The changes led to changes in receptor specificity, enhanced viral power and changes in resistance to amantadine ammonia. Phylogenetic tree analysis of HA genes showed that the strains from Yunnan, Sichuan and Guangzhou belonged to sub-branch 2.3.4.4 of H5 clade 2.3.4. Six strains of avian influenza virus A(H5N6) isolated from environmental samples and the two strains from patients in Yunnan province related to the strains from Guangzhou. All strains belonged to the same clade. The two strains from Yunnan showed a certain genetic distance from Sichuan and Guangzhou strains.ConclusionsThe genetic diversity and complexity of avian influenza A(H5N6) virus in Yunnan province are observed. The variation in functional loci of the virus may be one of the cross-species transmission mechanisms of of avian influenza virus. Dynamically monitoring the distribution and genetic variations of avian influenza viruses in live poultry markets should be used to provide research data for the study of cross-species transmission of avian influenza viruses
