MTHFR C677T基因多态性与原发性高血压相关性的荟萃分析 预览

Association of MTHFR C677T gene polymorphism with the risk of essential hypertension:A meta-analysis

摘要 目的探讨亚甲基四氢叶酸还原酶(MTHFR)C677T基因多态性与原发性高血压风险的关系。方法对电子数据库进行搜索,检索时限为数据库建库至2016年7月。数据由2位作者独立提取,同时对相关的数据库进行了检索和审查,采用Revman12.0软件进行分析。使用95%可信区间(95%CIs)的比值比(ORs)来评估MTHFR C677T多态性与高血压风险之间的关系。纳入C677T基因多态性研究的41项课题,其中包括9 050个病例和9 818个对照。结果荟萃分析结果表明,MTHFR C677T基因多态性增加了原发性高血压的风险。等位基因模型(T vs.C)、主导基因模型(TT+CTvs.CC)、隐性模型(CT+TT vs.CC)、纯合子模型(TT vs.CC)和杂合子模型(CT vs.CC)合并95%的可信区间分别为1.34(1.22~1.46)、1.39(1.26~1.53)、1.48(1.26~1.74)、1.70(1.42~2.03)和1.30(1.19~1.43)。通过Begg漏斗图和Egger检测,未发现明显的发表偏倚。结论本荟萃分析支持MTHFR C677T基因多态性与原发性高血压之间存在明显相关性。 Objective To explore the relationship between MTHFR (methylenetetrahydrofolate reductase)C677T gene polymorphism and the risk of essential hypertension (EH).Methods Electronic databases were searched to identify relevant studies published with retrieval time from their inception to July 2016.The data were extracted by two inde- pendent authors at the same time,and the related databases were searched,examined,and analyzed by Revman12.0 software.The 95%confidence interval (95%CI)Odds ratios (OR)was used to assess the relationship between MTHFR C677T polymorphism and the risk of hypertension.Finally,41 studies were included in the C677T gene polymorphism research,including 9 050 cases and 9 818 control groups.Results Results The results of the meta-analysis indicated that MTHFR C677T polymorphism contributed to an increased risk of EH.The pooled OR corresponding to 95%CI of the allele model (T vs.C),the dominant model (TT+CT vs.CC),the recessive model (TT vs.CT +CC),the homozygote model (Tr vs.CC),and heterozygote models(CT vs.CC)were 1.34 (1.22-1.46),1.39 (1.26-1.53 ),1.48 (1.26 -1.74),1.70 (1.42 -2.03 )and 1.30 (1.19 -1.43 ),respectively.No significant publication bias in the meta -analysis was found through Begg's funnel plot and Egger's test.Conclusions The present study supports the evidence for the association between the MTHFR C677T gene polymorphism and EH.
