钩藤提取物对MPTP致帕金森病小鼠的保护作用及机制的研究 被引量:4

Neuroprotective effect and mechanism of Uncaria rhynchophyUa (miq.) jacks extract against MPTP induced mouse model of Parkinson's disease

摘要 目的评价钩藤提取物的抗帕金森病活性及其可能的作用机制。方法经腹腔注射MPTP(1-甲基-4-苯基-1,2,3,6-四氢吡啶),建立小鼠帕金森病(PD)模型。观测小鼠的自主活动、爬杆能力、滚轴能力及悬挂能力等行为学表现,同时检测小鼠脑黑质中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性及丙二醛(MDA)水平的变化。结果经MPTP诱导的小鼠,其自主活动次数显著减少、爬杆时间差值显著增加、附着能力评分显著降低,钩藤提取物预处理后能显著增加小鼠在测试时间内的自发站立次数(P<0.01),显著缩短爬杆时间差值,提高附着能力分值(P<0.05),提高小鼠脑内SOD、GSH-Px水平(P<0.05),显著降低MDA水平明显下降(P<0.01)。结论钩藤提取物能提高PD小鼠肢体运动协调能力,可能通过抑制氧化损伤起到对PD模型小鼠起保护作用。 Objective To study the neuroprotective effect of Uncaria rhynchophylla( miq.) jacks extract against MPTP- induced mouse model of Parkinsons disease( PD) and its mechanism. Methods Mice PD model was established by intraperitoneal injection of MPTP( 1- methyl- 4phenyl- 1,2,3,6- terahydropyridine). Spontaneous movement,pole climbing ablility,rod rotation ablility and traetion ablility were observed. Detect the changes of SOD in substantia nigra,GSH- Px activity and levelof MDA simultaneously. Results Spontaneou movement frequency significantly decreased in mice induced by MPTP,time difference of pole climbing significantly incressed,and adhesive ability grade significantly decreased. Preteatment with Uncaria rhynchophylla( miq.)Jacks extract could significantly increase spontaneous stand- up times during test( P <0. 01),shortes the time difference of pole climbing,increase adhesive ability( P <0. 05),increase levels of SOD,GSH- Px in brain( P <0. 05),and decrease level of MDA( P <0. 01). Conclusion Uncaria rhynchohylla( miq.) Jacks can improve the sport coordinate ability of PD mice,and protect PD model mice by inhibiting the level of oxidative stress injury.
