二肽基肽酶6在难治性颞叶癫痫患者中的表达 认领

Altered expression of dipeptidyl peptidase 6 in patients w ith intractable temporal lobe epilepsy

摘要 目的 探讨难治性癫痫患者颞叶皮质中二肽基肽酶6(DPP6)的表达情况。方法 在重庆医科大学建立的脑组织库中,按照随机化原则抽取27例癫痫患者的脑组织(癫痫组)和15例脑外伤患者的脑组织(对照组),应用免疫组织化学染色、蛋白质印迹杂交、RT-PCR法检测DPP6的表达水平。结果 DPP6表达于两组大脑神经元的细胞膜,蛋白质印迹杂交结果显示,癫痫组和对照组DPP6的平均光密度(OD)值分别为0.86±0.04和0.37±0.02,差异有统计学意义(P〈0.001)。癫痫组和对照组脑组织DPP6 mRNA的相对表达量分别为0.74±0.03和0.35±0.03,差异有统计学意义(P〈0.001)。结论 异常表达的DPP6可能参与了难治性癫痫的形成。 Objective To investigate the expression of dipeptidyl peptidase 6( DPP6) in the neocortex of patients with intractable temporal lobe epilepsy( TLE) and discuss its role in the formation of intractable epilepsy. Methods DPP6 expressions were detected in 27 temporal neocortex tissue samples from patients with intractable TLE( epilepsy group)and 10 histologically normal temporal lobe tissue samples from patients without epilepsy( control group) by immunohistochemical staining,Western blotting and RT-PCR methods. Results DPP6 was found to be expressed mainly in the membrane of neurons. Western blotting analysis showed that the mean optical density( OD) ratios of DPP6 in the temporal neocortices of epilepsy group and control group were 0. 86 ± 0. 04 and 0. 37 ± 0. 02,respectively( P〈0. 001).The level of DPP6 mRNA in epilepsy group( 0. 74 ± 0. 03) was significantly higher than control group( 0. 35 ± 0. 03)( P〈0. 001). Conclusion The abnormal expression of DPP6 may contribute to epileptogenesis and represent a novel therapeutic target.
