目的 分析江苏省2014年流行性腮腺炎病毒（MuV）的基因特征。方法 采用Vero/SLAM细胞分离流行性腮腺炎病毒,采用RT-PCR法对25株MuV分离株的小疏水蛋白（Small Hydrophobic Protein,SH）基因片段进行扩增和序列测定,结合WHO MuV参考株进行分子流行病学研究。结果 通过比较核苷酸和氨基酸同源性和构建亲缘关系树发现,江苏省2014年25株MuV分离株同属F基因型,但序列间存在差异。核苷酸和氨基酸同源性分别为94.0%-100.0%和84.9%-100.0%。MuV分离株在SH基因编码的氨基酸保守位点也发生了变化。结论 江苏省2014年流行的MuV基因型同属F基因型,为F基因型MuV的多个传播链引起,但与F基因型参考株序列差异较大,发生了一定程度变异。
Objective To analyze molecular characteristics of mumps virus in Jiangsu Province in 2014. Methods Vero/ SLAM cells were used for mumps virus isolation. The SH gene fragment of 25 mumps virus isolates were amplified by RTPCR. PCR products were subjected to sequencing analysis and molecular epidemiological analysis combined with WHO mumps reference strains. Results Comparison of nucleic acids and amino acids homology and phylogenetic analysis showed that all 25 mumps virus strains in Jiangsu Province in 2014 were genotype F, with various sequences. The homology of SH gene fragment was 94.0%- 100.0% for nucleic acids and 84.9% - 100.0% for amino acids, respectively. The amino acids sequences of some conservative regions changed among mumps virus isolates. Conclusion Epidemic mumps virus in Jiangsu Province in 2014 were genotype F, which were involved in multiple transmission chains. The sequences were different compared to genotype F mumps virus reference strains, indicating certain genetic variation had occurred.
