β-谷甾醇对朱砂叶螨的触杀活性及杀螨机理初探 预览

The contact toxicity of β-sitosterol against Tetranychus cinnabarinus and its acaricidal mechanism

摘要 本研究旨在探究β-谷甾醇对朱砂叶螨的触杀活性及其对叶螨体态、内部结构的破坏。通过玻片浸渍法和叶片药膜法确定β-谷甾醇对朱砂叶螨杀螨的毒力,通过扫描电镜和透射电镜观察β-谷甾醇对螨亚显微结构的破坏,通过酶活检测方法,检测β-谷甾醇对乙酰胆碱酯酶和羧酸酯酶酶活的作用,通过荧光定量PCR方法测定β-谷甾醇对乙酰胆碱酯酶基因和羧酸酯酶基因相对表达量的影响。测定β-谷甾醇对朱砂叶螨雌成螨的LC30为1.0mg/mL。由扫描电镜和透射电镜观察可见,β-谷甾醇处理后朱砂叶螨表皮、肌纤维、线粒体、内质网、高尔基体等均受到不同程度的破坏。β-谷甾醇对朱砂叶螨乙酰胆碱酯酶基因表达量和酶活都没有影响。β-谷甾醇对解毒酶羧酸酯酶酶活没有直接影响,但诱导羧酸酯酶基因的表达。研究结果表明了β-谷甾醇可以有效杀死朱砂叶螨,并且对螨体造成不同程度的破坏,作为新型植物源农药具有一定的开发价值。证实乙酰胆碱酯酶不是β-谷甾醇的作用靶标,但β-谷甾醇处理会诱导螨体内解毒酶的表达。 This study is to investigate the acaricidal mechanism ofβ-sitosterol against Tetranychus cinnabarinus at cellular and molecular levels.The glass slide-dipping method and the leaf immersion method were used to measure the toxicity ofβ-sitosterol to mites.The scanning electron microscopy(SEM)and transmission electron microscopy(TEM)were used to observe the change of the submicroscopic structure of mites after exposure toβ-sitosterol.The enzyme activities tests were used to determine the effects ofβ-sitosterol on acetylcholinesterase and carboxylesterase.The effects ofβ-sitosterol on the expression of acetylcholinesterase and carboxylesterase genes were determined by quantitative PCR.The LC30 value ofβ-sitosterol on the female adult mites was determined as 1.0 mg/mL.SEM and TEM observations revealed thatβ-sitosterol treatment destroyed the mites' cell structure such as cuticle,muscular filament,mitochondria,endoplasmic reticulum and golgiapparatus.β-sitosterol had no effect on acetylcholinesterase activity and its gene expression.β-sitosterol has no direct interaction with the carboxylesterase,but can induce its expression.It was shown thatβ-sitosterol could effectively kill mites,and cause damage to mites' structure.It could be a new potential botanical acaricides.β-sitosterol did not directly interact with carboxylesterase and acetylcholinesterase,but it induced the expression of carboxylesterase gene.
