目的 观察补肾安胎冲剂对复发性自然流产（recurrent spontaneous abortion,RSA）小鼠胚胎丢失率和蜕膜组织血管内皮生长因子（vascular endothelial growth factor,VEGF）及其受体的影响。方法 采用Clark经典RSA小鼠模型,将RSA小鼠随机分为模型组、补肾安胎冲剂低剂量（11.7g/kg）组、补肾安胎冲剂高剂量（23.4g/kg）组,黄体酮胶囊（156mg/kg）组,另取正常妊娠小鼠作为正常组,每组8只。末次给药后,处死小鼠,计算胚胎丢失率;光镜下观察蜕膜组织血管形态学改变;采用免疫组化法检测蜕膜组织VEGF、血管内皮生长因子受体2（vascular endothelial growth factor receptor 2,VEGFR-2）、可溶性血管内皮生长因子受体-1（soluble vascular endothelial growth factor receptor 1,sVEGFR-1）的表达;采用RT-PCR法检测小鼠蜕膜组织中VEGF、VEGFR-2及sVEGFR-1mRNA的表达。结果 补肾安胎冲剂能改善蜕膜组织血管形态学变化,升高蜕膜组织中VEGF和VEGFR-2蛋白含量,降低sVEGFR-1蛋白含量;能显著降低RSA小鼠胚胎丢失率（P〈0.05）,升高蜕膜组织中VEGF和VEGFR-2mRNA表达水平,降低sVEGFR-1mRNA表达水平（P〈0.05）,呈现明显的量效关系。结论 补肾安胎冲剂对RSA小鼠具有一定的保护作用,其机制可能与调控VEGF及其受体,改善蜕膜组织血管重铸有关。
Objective To observe the effects of Granules for Tonifying the Kidney and Preventing Miscar- riage（GTKPM）on embryo loss rate and decidual vascular endothelial growth factor （VEGF） and its recep- tor in mice with recurrent spontaneous abortion （RSA）. Methods The classical Clark RSA mice were se- lected and randomly divided into model group, low-dose GTKPM group （11.7 g/kg）, high-dose GTKPM group （23.4 g/kg）, and progesterone capsule group （156 mg/kg）, each with 8 mice. Another 8 normal pregnancy mice were selected as the control group. After the last administration, the mice were sacrificed, and the embryo loss rate （ELR） was calculated. The changes in the vascular morphology of decidual tissue were observed under a light microscope. The protein expression of vascular endothelial growth factor （VEGF）, vascular endothelial growth factor receptor 2 （VEGFR-2）, and soluble vascular endothelial growth factor receptor 1 （sVEGFR-1） in decidual tissue was measured by immunohistoehemistry. Moreover, RT-PCR was performed to measure the mRNA expression of VEGF, VEGFR-2, and sVEGFR-1 in decidual tissue. Results GTKPM improved the vascular morphology of decidual tissue, increased the pro- tein expression of VEGF and VEGFR-2 in decidual tissue, and reduced the protein expression of sVEGFR- 1. In addition, GTKPM significantly reduced the ELR of RSA mice （P〈0.05）, increased the mRNA ex- pression of VEGF and VEGFR-2 in decidual tissue, and reduced the mRNA expression of sVEGFR-1 （P〈0.05）,with a significant dose-effect relationship. Conclusion GTKPM has a certain protective effect on RSA mice, which may be related to its regulatory effect on VEGF and its receptor and improving the vascular remodeling of decidual tissue.
