The expression of Wnt/β-Catenin in the brain of Alzheimer's disease rats through bone marrow stromal stem cells for the treatment

摘要 目的探讨BMSCs移植对AD模型大鼠的治疗作用。方法将大鼠随机分成5组,正常组、模型组、模型对照组、治疗组、治疗对照组,每组10只。双侧海马区注射Aβ23-35建立AD大鼠模型后,治疗组进行BMSCs的移植治疗。治疗对照组用氢溴酸加兰他敏进行灌胃治疗。结果行为学实验检测各组大鼠平均逃避潜伏期及首次跨越原平台时间,模型组与正常组比较(P〈0.01),治疗组与治疗对照组与模型组比较(P〈0.01),与正常组比较(P〈0.05),均有显著性差异;形态学实验HE染色显示模型组颗粒细胞及锥体细胞有破坏现象,治疗组和治疗对照组明显恢复。免疫组化检测显示正常组和模型对照组可见海马区阳性蛋白表达较多而模型组阳性表达明显减少,治疗组和治疗对照组阳性表达较模型组明显增多。结论 BMSCs移植治疗AD的作用机制可能与通过调节Wnt信号转导通路,影响了学习记忆相关酶及蛋白在海马区的表达而发挥作用有关。 Objective To investigate the therapeutic effect of BMSCs transplant on AD model rats. Methods Rats were randomly divided into five groups,the normal group,model group,model control group,the treatment group,the treatment control group,n=10.After bilateral hippocampus injection Aβ23-35,The rat model of AD was established,BMSCs transplantation therapy in the treatment group.We performed gavage treatment with galantamine hydrobromide in the treatment control group. Results Behavioral experiments detected the average escape latency of rats and the first time across the original platform,Model group compared with normal group(P〈0.01),treatment group and the treatment group compared with model group(P〈0.01),compared with normal group(P〈0.05),There were significant differences.Experimental Morphology HE staining showed that the model group granule cells and pyramidal cells have damage phenomenon,treatment group and control group obviously recovery.Immunohistochemistry showed that normal group and model control group showed more positive protein expression in the hippocampus,however the expression in the model group was significantly reduced.Treatment group and treatment group expression was significantly increased compared with the model group. Conclusion BMSCs transplantation treated AD which possible mechanism of action by adjusting the Wnt signaling pathway,affect learning and memory-related enzyme protein expression in the hippocampus play a role related.The expression of enzymes and proteins associated learning and memory in the hippocampus which play an important role.
