热毒血瘀证结肠癌荷瘤小鼠模型的建立及清热解毒活血化瘀法对其的影响 被引量:1

Development of mice model of colorectal tumor-bearing with syndrome of heat-toxicity and blood stasis and the effects of removing toxicity and activating blood and resolving stasis therapy

摘要 目的:建立热毒血瘀证荷结肠癌复合动物模型并探索清热解毒活血化瘀法对其影响。方法:用脂多糖(LPS)尾静脉和腹腔注射建立热毒血瘀证模型,C26皮下注射建立荷瘤模型,设空白对照组,单纯荷瘤组,复合模型对照组,复合模型高、中、低剂量组,中药干预后行量化评分表、局部微循环血流量及血液流变学相关指标的检测。结果:复合模型各组造模1周起出现不同程度热毒血瘀证。与复合模型对照组比较,复合模型高、中剂量组造模12-42d量化评分值明显减轻(P<0.05,P<0.01)。造模后复合模型组局部血流量减低,给药后各剂量组局部血流量增高,以复合模型高剂量组更为明显。造模后,各组全血黏度值均升高明显,运用活血化瘀法后可在一定程度上改善血液的高黏滞状态。结论:本实验建立热毒血瘀证结肠癌的病证复合模型,符合中医证型特点;采用不同剂量的清热解毒活血化瘀中药进行干预,能不同程度减少热毒血瘀的症状和体征。 Objective: To develop the mice model of colorectal tumor-bearing with syndrome of heat-toxicity and blood stasis and observe the effects of activating blood and resolving stasis therapy. Methods: Establishing model by LPS of tail intravenous injection and peritoneal injection and C26 tumor cells subcutaneous injection, mice were randomly divided into 6 groups, respectively called blank control group(group A), tumor-bearing mice group(group B), control group with tumor(group C), tumor-bearing mice group with high dose herb medicine(group D), tumor-bearing mice group with middle dose herb medicine(group E), tumor-bearing mice group with low dose herb medicine(group F). After drug administration, quantified scoring, local microcirculation blood flow and hemorheology indices before and after molding were set up. Results: At the first week, mice in group of disease with TCM zheng showed different kinds of rise in temperature as well as vein stasis syndrome, mice in group D and group E had a gradually decreased score from molding 12 days to 42 days(P<0.05, P<0.01). After molding, regional blood flow of mice in the groups of disease integrating with TCM zheng reduced, after administration, regional blood flow of mice in these groups increased, especially in group D. After molding, there was significant increasing in the whole blood viscosity. Activating blood circulation can improve the state of hypercoagulability of blood to some extent. Conclusion: In this current study, we successfully developed the mice model of colorectal tumor-bearing with syndrome of heat-toxicity and blood stasis that well conformed to the TCM syndrome characteristics. In addition, the treatment of herbs with activating blood and resolving stasis at different doses improved the symptoms and signs to some extent.
