植物油降解细菌的培养条件优化和分离鉴定 预览

摘要 m从自然环境中筛选出的高效油脂降解菌,可应用于对油脂废水的处理.然而,碳源、氮源、微量元素等营养物质对微生物的生长、繁殖和代谢有很大影响.以大豆油为唯一的碳源,重点研究微生物降解植物油废水的最佳营养条件,分离植物油降解菌菌种.通过16SrDNA扩增和基因测序对其进行鉴定.结果表明,复合菌群作用的最佳营养条件为:氮源种类为氯化铵(NH4Cl),优水平浓度为2.108g/L,C/N 4.8;磷源种类为磷酸二氢钾(KH2PO4),优水平浓度为1.3g/L,微量元素配方及优水平浓度为七水合硫酸镁(MgSO4·7H2O,AR)0.08g/L,七水合硫酸亚铁(FeSO4·7H2O,AR)0.16g/L,五水合硫酸铜(CuSO4·5H2O,AR)0.08g/L,无水氯化钙(CaCl2,AR)0.008g/L,乙二胺四乙酸二钠(C10H14N2Na2O8·2H2O,AR)0.008g/L,七水合硫酸锌(ZnSO4·7H2O,AR)0.08g/L.16SrRNA序列分析法鉴定其中1株菌株为Bacillus cereus ATCC 14579,隶属于厚壁菌门,最适生长温度在20-45℃. The efficient oil-degrading strain which is from the natural environment can be applied to treating oil wastewater. Carbon source, nitrogen source, trace elements and other nutrients have a great effect on the growth, reproduction and metabolism of microorganisms. This study aims at searching for the best nutritional condition of the oil-degradation strain treating oil wastewater with soybean oil as the sole carbon source and separating oil-degradation straia DNA is extracted, and PCR is used to amplify 16S rDNA The results show that the optimum nitrogen source is NH4C1, whose optimal concentration is 2. 108 g /L,C/N is 4. 8; the optimum phosphorus source is KH2P04,whose optimal concentration is 1. 3 g /L ,the optimum microelement’ s formula is 0. 08 g/ L MgSO4 , 7H2O,0.16 g/L FeSO4 , 7H20 ,0. 08 g/L CuS04 , 5H2O,0. 008 g/L CaCl2,0. 008 g/L C10H14N2Na2O8 , 2H20 , 0. 08 g/L ZnS04 , 7H20. A strain with soybean oil as sole carbon source and energy is identified as Bacillus cereus ATCC 14579, which is affiliated with phylum fir- micute, and the optimum growth temperature is within the range from 20℃ to 45℃.
