中国2型糖尿病人群葡萄糖依赖性促胰岛素肽受体基因多态性与冠心病风险的相关性研究 被引量:5

Association of genetic variability in glucose-dependent insulinotropic polypeptide receptor and coronary artery disease in Chinese Han type 2 diabetic patients

摘要 目的观察中国汉族T2DM患者中葡萄糖依赖性促胰岛素肽(GIP)及其受体(葡萄糖依赖性促胰岛素肽受体,GIPR)基因多态性与冠心病风险的相关性。方法纳入666例T2DM患者,其中冠心病组390例,对照组276例。根据HapMapCHB数据库选取单体型标记的单核苷酸多态性(tagSNPs)。GIPR基因共4个tag-SNPs,分别为rs1800437、rs11671664、rs4803846和rs12709891。应用聚合酶链式反应-限制性内切酶片断长度多态性方法,对GIPR基因tag-SNPs进行等位基因和基因型频率分析,研究GIPR基因多态性与冠心病风险的关系。结果两组GIPR基因SNPs等位基因频率比较,差异无统计学意义。冠心病组和对照组rs1800437、rs11671664、rs4803846和rs12709891等位基因频率分别为79.8%vs 82.5%,62.6%vs 59.3%,89.0%vs 89.3%和48.8%vs 50.4%。两组GIPR基因rs1800437和rs4803846单体型组合比较,差异无统计学意义。冠心病组和对照组单体型组合GG、GC和AG频率分别为69.2%vs 72.0%,19.8%vs 17.3%和10.6%vs 10.5%。结论中国T2DM患者中,未发现GIPR基因多态性与冠心病发生风险相关的SNPs,提示GIP受体基因变异可能不是中国T2DM患者冠心病风险的主要决定因素。 Objective To investigate the relationship between glucose-dependent insulinotropic polypeptide receptor(GIPR)gene polymorphisms and the risk of cardiovascular disease in Chinese Han type 2diabetic patients(T2DM). Methods Of total 666 T2 DM patients,390 with coronary artery disease(CAD)as cases and 276 without CAD as controls were recruited.We selected and genotyped four haplotype-tagging single nucleotide polymorphisms of GIPR gene(rs1800437,rs11671664,rs4803846 and rs12709891).The four tag-SNPs were selected from CHB data in HapMap Phase II database(r2〈0.8and MAF≥0.05,R#27).The PCR-RFLP assay was adopted to genotype haplotype-tagging single nucleotide polymorphisms(SNPs)in GIPR gene.Allele and genotype frequencies were compared between the CADpositive and controls in all SNPs. Results(1)There was no statistical difference in the distribution of allele frequencies between the cases and controls in GIPR gene(rs1800437,79.8%vs 82.5%;rs11671664,62.6%vs 59.3%;rs4803846,89.0% vs 89.3%;rs12709891,48.8% vs 50.4%;respectively,P〉0.05).(2)There were no significant differences in the distribution of GIPR rs1800437+rs4803846haplotype between two groups(GG,69.2% vs 72.0%;GC,19.8% vs 17.3%;AG,10.6% vs 10.5%;respectively,P〉0.05). Conclusion GIPR gene mutation may not be a decisive factor of coronary heart disease in type 2diabetic population in China.
