Objective To analyze the genetic characteristics of rubella virus（ RV） strains isolated in Jiangsu province from2013 to 2015. Methods Rubella virus strains were isolated on Vero-Slam cells from the throat swabs samples of the suspected patients. The fragments containing 739 nucleotides of E1 genes from 7 rubella virus strains were amplified by RT-PCR method,the PCR products were directly sequenced and analyzed and WHO rubella virus reference strains were conducted for epidemiologic study. Results 3 isolated strains were closely related to reference strains of 1 E genotypes,the homology of nucleotide and amino acid were within 99. 2%-100. 0% and 99. 6%-100. 0%,respectively. Four isolated strains were closely related to reference strains of 2 B genotypes,the homology of nucleotide and amino acid were within 98. 9%-100. 0% and100. 0%,respectively. No variation was detected at hemagglutination inhition and neutralization sites. Two amino acid mutation from Leu to Phe in the aa338 amino acid and from Leu to Val in the aa357 amino acid of E1 protein was identified,respectively.Conclusion Genotype 1 E and 2 B of rubella virus were the predominant genotypes in Jiangsu province during 2013-2015,which was similar to the other provinces in China. The amino acid sequence of E1 protein was highly conservative.
