氟伏沙明抗抑郁作用与用药时间的相关性研究 被引量:1

Experimental Study on the Relationship Between the Antidepressant Effect of Fluvoxamine and Medication Time

摘要 目的探讨氟伏沙明的抗抑郁作用与用药时间的相关性。方法取雄性ICR小鼠77只随机排号,1-ll号为对照组,12-77号为观察组。将观察组又随机分为6个亚组(每组各11只)。6个亚组分别在9:00(T1)、13:00(T2)、17:00(T3)、21:00(T4)、1:00(T5)、5:00(T6)时均给予氟伏沙明片(200mg溶于100mL水中)灌胃,每天1次。对照组给予同等体积的生理盐水灌胃.每天1次。连用7d后检测不动累积时间(5rain内)。另取雄性ICR小鼠36只,随机分为观察组和对照组各18只.各组叉随机分为明期组、暗期组各9只。明期组在同日内9:00给予氟伏沙明,暗期组在同日21:00给予氟伏沙明(用法用量同观察组)。连用7d后.将小鼠放于暗箱中,检测小鼠自发性活动(10min内)。结果观察组各时间点不动累积时间低于对照组,差异有统计学意义(P〈0.05);观察组在T4时不动累积时间低于其他时间点,差异有统计学意义(P〈0.05);观察组、对照组中的暗期组的运动距离比明期组长,差异有统计学意义(P〈0.05);观察组中明期、暗期组的运动距离、静止时间和对照组的明期、暗期组比较,差异无统计学意义(P〉0.05)。结论氟伏沙明在小鼠中抗抑郁作用的强弱与用药时间有关,具有昼夜节律性。 OBJECTIVE To explore the relationship between the antidepressant effect of fluvoxamine and medication time. METHODS 77 male ICR mice were randomly arranging, 1-11 number was the control group, 12-77 number was the observation group.The observation group was randomly divided into 6 sub groups (each group was 11 ), 6 sub groups respectively in 9:00 (T1), 13:00(T2) , 17:00 (T3) , 21:00 (I'4) , 1:00 (T5) , 5:00 (T6) were given fluvoxamine (200 mg tablets dissolved in 100 mL water) by gavage, 1/d. The control group was given the same volume of saline, 1/d.After the 7 d was used, the cumulative time (5 min) was measured .Another 36 male ICR mice were used, they were randomly divided into observation group and control group, 18 in each group, each group was randomly divided into light group and dark group, 9 in each group.Light group 9:00 was given fluvoxamine on the same day, the dark group was given fluvoxamine on the same day 21:00( usage and dosage were the same as those in the observation group). After using 7 d, put the mouse in the black box, the spontaneous activity of mice was detected ( in 10 min). RESULTS The observation group was significantly lower than the control group (P〈0.05). In the observation group, the cumulative time in T4 was significantly lower than other time points ( P〈0.05 ). Motion distance comparison, the observation group and the control group in the dark group moving distance was longer than light group (P〈0.05). The observation group in the period of moving distance of dark group, light group and the control group at the time of the dark group were compared, the difference was not significant (P〉 0.05). CONCLUSION Fluvoxamine antidepressant effect strength in mice is associated with medication time,it has circadian rhythm.
