二补助育汤对胚胎着床障碍小鼠胚胎着床数目及M-CSFmRNA表达的影响 被引量:1

Influence of Erbu Zhuyu Decoction on nidation number and M-CSF mRNA expression in mouse model of embryo implantation dysfunction

摘要 目的:探讨二补助育汤对胚胎着床障碍模型小鼠胚胎着床率及子宫内膜巨噬细胞集落刺激因子(M-CSF)mRNA表达的影响。方法:将112只C57小鼠随机分为空白组,模型组,补佳乐组,阿司匹林组,中药低、中、高剂量组,每组16只。用吲哚美辛建立胚胎着床障碍模型,予以相应药物灌胃,小鼠妊娠第5天下午脱颈处死,检测各组妊娠率、平均着床位点数、子宫内膜M-CSF mRNA表达量。结果:(1)裸眼观察小鼠子宫:空白组宫体红润,着床处充血明显;模型组宫体苍白、细长;用药各组较空白组宫体苍白、着床位点分布稀疏不均匀。(2)妊娠率及平均着床位点数:与空白组比较,模型组平均着床位点数显著降低(P〈0.05);中药中、高剂量组平均着床位点数较模型组显著增高(P〈0.05)。(3)M-CSF mRNA表达:模型组小鼠子宫M-CSF mRNA表达较空白组显著降低(P〈0.05);与模型组比较,中药各剂量组和西药各组M-CSF mRNA表达偏高,其中中药中剂量组显著增高(P〈0.05);中药中剂量组小鼠子宫M-CSF mRNA表达高于西药各组(P〈0.05),高于中药低、高剂量组,但无显著差异。结论:二补助育汤可能通过增加M-CSF mRNA表达,从而改善子宫内膜容受性,利于胚胎着床。 Objective:To explore the effects of Erbu Zhuyu Decoction on blastocyst implantation rate and M-CSF mRNA expression in mouse model of embryo implantation dysfunction.Methods:A total of 112C57 mice were randomly divided into seven groups:blank group,model group,Progynova group,Aspirin group,and low-,middle-,high-dose group of Erbu Zhuyu Decoction,16 mice in each group.Embryo implantation dysfunction model was established by indomethacin.These mice were given corresponding drugs to the stomach and then were taken off the neck to death on the 5 th day of pregnancy.Those groups' blastocyst implanted number and M-CSF mRNA expression were detected.Results:(1)For visual observation of mice's uteri:the uteri of blank group were rosy and had congestive significantly sites; uteri of model group were pale and slender; traditional Chinese medicine(TCM) in each dose group and Western medicine groups were pale compared with blank group,and distribution of implantation sites were sparse and asymmetry.(2)For pregnancy rate and average implantation number:model group was worse than blank group(P〈0.05); middle-dose group and high-dose group were better than model group(P〈0.05).(3)For M-CSF mRNA expression:model group was significantly lower than blank group(P〈0.05); compared with the model group,the expression of M-CSF mRNA in western medicine groups and the Erbu Zhuyu Decoction groups increased; the expression of M-CSF mRNA in middle-dose group significantly increased compared with blank group and Western medicine groups(P〈0.05); the expression of M-CSF mRNA in middle-dose group was higher than low-dose group and high-dose group,but there was no significant difference.Conclusion:Erbu Zhuyu Decoction may improve endometrial receptivity by increasing M-CSF mRNA expression,which is conductive to embryo implantation.
