目的研究芦荟大黄素对高脂血症大鼠心肌炎的作用。方法采用高脂饮食饲养Wistar雄性大鼠4周,4周后检测血清血脂指标,确定高脂血症模型成功建立后,灌胃给予芦荟大黄素（100 mg/kg）6周,6周后取心脏组织,应用Real-time PCR技术测定miRNA-33的表达水平,应用Western blot方法检测炎性细胞因子IL-1β蛋白表达水平。结果 （1）与对照组比较,高脂饮食喂养四周后的大鼠血清中TC,LDL-c含量显著升高（P〈0.001）,TG含量显著升高（P〈0.05）,HDL-c含量无显著差异（P〉0.05）;（2）与对照组相比,高脂血症大鼠心脏组织中miR-33表达显著增加（P〈0.001）,给予芦荟大黄素后,miRNA-33表达水平显著降低（P〈0.001）;（3）与对照组相比,高脂血症大鼠心脏组织中IL-1β蛋白表达明显增加（P〈0.001）,给予芦荟大黄素后IL-1β蛋白表达显著降低（P〈0.001）。结论芦荟大黄素通过抑制高脂血症大鼠心脏中miRNA-33的表达和炎性因子IL-1β的产生减轻心脏炎症反应。
Objective To investigate the effect of Aloe-emodin on inflammatory response in hyperlipidemia rat hearts. Methods Male Wistar rats were fed with high fat diet for four weeks.Serum lipid indexes were detected to validate hyperlipidemia model. After modeling,a part of hyperlipidemia rats were intragastrically administrated with Aloe-emodin（100 mg/kg） for six weeks. Subsequently,all rat hearts were collected. The level of miRNA-33 was detected by Real-time PCR and protein expression of inflammation factor IL-1β was detected by Western blot. Results（1）After feeding with high fat diet for four weeks,the serum lipid contents of TC,TG and LDL-c were significantly increased compared with control group,while no significant change of HDL-c was found;（2）The level of miRNA-33 was markedly higher in hyperlipidemic rat hearts than sham rats,which was inhibited by Aloe-emodin;（3）The level of inflammation factor IL-1β was significantly increased in hyperlipidemic rat hearts compared with sham rats,which was reduced by Aloe-emodin. Conclusion Aloe-emodin inhibits miRNA-33 expression and inflammatory factor IL-1β production to attenuate rat heart inflammatory response against hyperlipidemia.
