穿心莲内酯对糖尿病肾病小鼠的治疗作用及机制 预览 被引量:1

Effects and mechanism ofandrographolide on diabetic nephropathy mice

摘要 利用自发2型糖尿病基因工程Leprdb/db小鼠糖尿病小鼠模型,探讨穿心莲内酯(andrographolide,Andro)对糖尿病肾病的作用及机制。利用试剂盒检测Andro治疗Leprdb/db小鼠24h后的尿蛋白、血肌酐和血尿素氮水平;利用HE染色和PAS染色分别检测Andro对糖尿病小鼠肾脏组织的病理改变和还原糖水平的影响;采用免疫组织化学染色法检测Andro对Leprdb/db小鼠肾脏的纤维粘连蛋白(FN)和NOX-4的影响。结果显示,Andro治疗组(1mg/kg,ip)的24h后Leprdb/db小鼠的尿蛋白、血肌酐和血尿素氮等生化指标水平明显低于对照组,Andro能抑制Leprdb/db小鼠肾脏的病理改变和FN、NOX-4的表达,表明Andro可以显著改善自发2型糖尿病基因工程小鼠Leprdb/db小鼠的糖尿病肾病。 To investigate the effects and mechanism of andrographolide(Andro)on type II diabetic nephropathy in Lepr db/db mice,levels of urinary protein,serum creatinine and blood urea nitrogen after24h of Andro(1mg/kg,ip)treatment on Lepr db/db mice were measured by kit.The changes on renal pathology and sugar levels in diabetic mice treated by Andro were observed by HE staining and PAS staining.The expression of fibronectin(FN)and NOX-4protein were analyzed by immunofluorescence.Results showed that levels of urinary protein,serum creatinine and blood urea nitrogen in Lepr db/db mice after Andro treatment for24h were significantly lower than those of the control group.Kidney pathologic change was inhibited,and FN and NOX-4expression were markedly decrease in the Andro group.In conclusion,Andro can significantly reduce the diabetic nephropathy in Lepr db/db mice with spontaneous type II diabetes mellitus.
