江西和广东烟草青枯菌对噬菌体的敏感性及聚类分析 预览

Cluster Analysis of Bacteriophage Sensibility of Ralstonia solanacearum from Guangdong and Jiangxi Provinces

摘要 为探索不同噬菌体对不同烟草青枯菌的侵染能力,选择从土壤中分离的6个青枯菌噬菌体,通过双层平板噬菌斑检测法,测定6个噬菌体P1521、P1553、P1556-1、P1556-2、P574-2和P7-1对江西和广东两省烟区75个烟草青枯菌的侵染和裂解情况,并基于青枯菌对3个噬菌体的敏感性差异,进一步对75个青枯菌进行聚类分析。结果表明:6个噬菌体分别可侵染65、63、61、57、57和24个烟草青枯菌菌株,噬菌体P1556-1和P1521寄主范围较广,噬菌体P1553寄主范围较窄;基于P1556-1、P574-2和P1553等3个噬菌体,对75个青枯菌进行聚类分析,将75个烟草青枯菌分为7个遗传组(Rs-I~Rs-VII),其中Rs-IV菌株数量较多,包括31个青枯菌,占供试菌株的41.33%,这些菌株对P1553噬菌体均表现不敏感,而对其它2个噬菌体敏感。 To investigate the infection of phages to of R.solanacearum,six phages named P1521,P1553,P1556-1,P1556-2,P574-2 and P7-1 isolated from soils were used to detect the infection and cleavage to 75 strains of Ralstonia solanacearum from Jiangxi and Guangdong provinces by double plate plaque assay.And a clustering analysis of 75 strains was carried out based on the sensitivity to phages.The results shows that six phages could respectively infect 65,63,61,57,57 and 24 of R.solanacearum strains.And the phage of P1556-1 and P1521 had a wide range of host,while the host range of the phage P1553 was narrow.The clustering analysis based on the infection by phages of P1556-1,P574-2 and P1553 showed that 75 strains of R.solanacearum were clustered into 7 genetic groups of Rs-I to Rs-VII.The number of R.solanacearum strain in group of Rs-IV was larger than 31,which accounting for 41.33%of the tested strains,and these strains were all resistant to phage of P1553 but sensitive to the phages of P1556-1 and P574-2.
