转化生长因子-β诱导视网膜色素上皮细胞增殖中NLRP3炎症小体的表达及意义 预览

Expression and significance of NOD-likereceptor 3 inflammasome in retinal pigment epithelial cell proliferation induced by transforming growth factor beta

摘要 目的探讨转化生长因子-β(transforming growth factor-β,TGF-β)刺激下Nod样受体蛋白3(nod-like receptor protein 3,NLRP3)炎症小体在视网膜色素上皮细胞(retinal pigment epithelium,RPE)增殖中的表达变化及意义。方法利用TGF-β刺激RPE细胞增殖模拟增生性玻璃体视网膜病变模型,随机分为空白对照组(Sham组)和TGF-β诱导RPE细胞增殖组(TGF-β组),TGF-β组再根据浓度(0.5 μg·L^-1 、2.5 μg·L^-1 、10.0 μg·L^-1 、12.5 μg·L^-1 )分组。利用MTT 微量酶比色法检测RPE细胞增殖情况;Western blot观察NLRP3蛋白的表达变化;利用标准ELISA试剂盒检测白细胞介素(interleukin,IL)-1β和 IL-18表达情况。结果与Sham组比较,0.5 μg·L^-1 TGF-β组RPE细胞增殖不明显,差异无统计学意义( P >0.05),但NLRP3、IL-1β和 IL-18表达明显提高(均为 P <0.01);2.5 μg·L^-1 、10.0 μg·L^-1 和12.5 μg·L^-1 TGF-β组RPE细胞增殖明显,NLRP3、IL-1β和 IL-18表达均提高(均为 P <0.05);与0.5 μg·L^-1 TGF-β组相比,2.5 μg·L^-1 、10.0 μg·L^-1 和12.5 μg·L^-1 TGF-β组RPE细胞增殖显著增加,NLRP3、IL-1β和 IL-18表达均下降,差异均有统计学意义(均为 P <0.05)。结论不同浓度TGF-β诱导RPE细胞增殖中NLRP3炎症小体表达变化不同,两者可能相互调控影响RPE细胞增殖,在增生性玻璃体视网膜疾病发生发展中具有重要的作用。 Objective To investigate the expression of NOD-likereceptor 3 (NLRP3) inflammasome in the proliferation of retinal pigment epithelial cells (RPE) by transforming growth factor beta (TGF-β) stimulation. Methods A proliferative vitreoretinopathy model was simulated by TGF-β stimulating RPE cells,which were randomly divided into two groups:blank control group (Sham group) and RPE cells proliferation stimulated by TGF-β group (TGF-β group).Then according to the different concentration of TGF-β,RPE cells were divided of 0.5 μg·L^-1 ,2.5 μg· L^-1 ,10.0 μg·L^-1 ,12.5 μg·L^-1 subgroups,respectively.The proliferation of RPE cells was detected by MTT Microenzyme colorimetry,and the expression of NLRP3 was observed by Western blot.Standard ELISA kit was applied to detect the expression of IL-1β and IL-18. Results Compared the with sham group,the proliferation of RPE was not obvious ( P > 0.05),but NLRP3,IL-1β and IL-18 expression was up-regulated significantly (all P < 0.01) in 0.5 μg·L^-1 TGF-β group;the proliferation of RPE cells,and expression of NLRP3,IL-1β and IL-18 were all increased in 2.5 μg·L^-1 ,10.0 μg·L^-1 ,12.5 μg·L^-1 groups (all P < 0.05).Compared with 0.5 μg·L^-1 TGF-β group,RPE cell proliferation was increased significantly ( P <0.05),but the expression of NLRP3,IL-1β and IL-18 were increased in 2.5 μg·L^-1 ,10.0 μg·L^-1 ,12.5 μg·L^-1 groups (all P <0.05). Conclusion Different concentrations of TGF-β induce different expression levels of NLRP3 inflammasome in RPE cell proliferation.They both play an important role in the occurrence and development of proliferative vitreoretinopathy by influencing each other and regulating RPE cell proliferation through different molecular mechanisms.
