枣疯病植原体保存体系构建 预览

Establishment of the in vitro culture system to preserve Candidatus Phytoplasma ziziphi

摘要 为建立枣疯病植原体组培苗保存体系,对比了不同外植体来源、培养基不同植物生长调节剂组合和质量浓度对枣疯病苗启动培养、继代培养、生根和植株体内植原体是否能够保持的影响.结果表明:休眠枝条水培芽比大田带病茎段启动培养成活率高,启动培养基中最适的生长调节剂组合为MS+KT2.0mg/L+NAA0.1mg/L,增殖培养基的最适组合为MS+6-BA2.0mg/L+NAA0.3mg/L,壮苗培养基为MS+KT2.0mg/L+NAA0.3mg/L,生根培养基为1/2MS+NAA0.5mg/L.对继代3~5次的组培病苗中植原体进行PCR检测表明,植原体稳定寄生在再生苗体内.建立的植原体组培苗保存体系,为开展枣疯病发生和防治机理的研究提供了技术支持. In order to establish a tissue culture system to preserve Candidatus Phytoplasma ziziphi,which is the pathogenic bacteria caused of gray jujube(Ziziphus jujuba ‘huizao’) witches’ broom disease.The effects of different explants sources,different plant growth regulator combinations and concentrations on initial culture,subculture and rooting culture were compared.The results showed that water-culture dormant buds have higher survival rate than that of field gathered diseased shoots.The optimal growth regulator combination in the initial medium was MS+KT2.0 mg/L+NAA 0.1 mg/L.The optimal combination of proliferation medium was MS+6-BA 2.0 mg/L+NAA 0.3 mg/L.To get strong seedling,the medium was MS+KT 2.0 mg/L+NAA 0.3 mg/L.And for rooting,the medium was 1/2 MS+NAA 0.5 mg/L.After 3 to 5 times subculture,the results of PCR detection by detecting 16 SrDNA in Candidatus Phytoplasma ziziphi indicated that the phytoplasma was still stably parasitic in the seedlings.The established tissue culture system of witches’ broom diseased jujube seedling could provide material for the study on the mechanism and control of phytoplasma disease.
