小檗碱对高糖高脂饲料联合链脲佐菌素诱导的糖尿病心肌病大鼠的保护作用 预览

The protective effect of berberine on diabetic cardiomyopathy rats induced by STZ and high-glucose-high-fat diet

摘要 目的 观察小檗碱(BBR)干预对糖尿病心肌病大鼠心肌以及心脏功能的影响。方法 建立高糖高脂饲料联合链脲佐菌素(STZ)诱导的2型糖尿病心肌病大鼠模型,另设健康对照组(NC),模型大鼠按照随机数字表法分为糖尿病心肌病组(DCM),小檗碱组(BBR100mg/kg、200mg/kg),二甲双胍组(二甲双胍200mg/kg),曲美他嗪组(曲美他嗪10mg/kg)。NC组和DCM组灌胃等量的溶媒,其他组灌胃相应剂量的该药物,持续12周,超声检测大鼠的左心室射血分数(LVEF),短轴收缩率(FS),E/A比值,全自动生化分析仪检测大鼠血清中的空腹血糖(FBG),三酰甘油(TG),胆固醇(TC),肌酸激酶(CK),乳酸脱氢酶(LDH)和天冬氨酸氨基转移酶(AST)的含量,计算心脏质量/体质量值(HW/BW),HE染色和Masson染色观察大鼠心肌的组织形态变化。结果 与NC组相比,DCM组大鼠血清FBG,TG,TC,CK,LDH,AST含量明显增加(P<0.05),心脏质量与体质量比增加(P<0.05),心脏FS和E/A(P<0.05)值明显下降,LVEF降低(P<0.05),HE染色可观察到DCM组心肌纤维断裂,排列紊乱,Masson染色则显示出DCM组胶原纤维含量增加;与DCM组比较,BBR高剂量组FBG,TG,CK,LDH,AST含量均下降(P<0.05),TC含量和心脏质量与体质量比降低(P<0.05),心脏的LVEF和FS值(P<0.05)增加,E/A也增加(P<0.05),心肌损伤即纤维化程度较DCM组轻。结论 小檗碱对2型糖尿病心肌病大鼠心肌结构和心脏功能的损伤具有保护作用。 Objective To observe the influence of berberine on myocardial and cardiac function in diabetic cardiomyopathy rats.Methods A rat model of type 2 diabetes cardiomyopathy induced by high-sugar and high-fat diet combined with streptozotocin (STZ) was established,and a normal control group (NC) was set up.The model rats were randomly divided into diabetic cardiomyopathy (DCM),Berberine group(BBR 100 mg/kg,BBR 200 mg/kg),Metformin group (Metformin 200 mg/kg),Trimetazidine group (TMZ 10 mg/kg).NC group and DCM group were given the same amount of solvent by gavage,while other groups were given the same dose of the drug which name indicated by gavage for 12 weeks.Left ventricular ejection fraction (LVEF),short axis systolic rate (FS),E/A ratio were measured by ultrasonography.The contents of fasting blood glucose (FBG),triglyceride (TG),cholesterol (TC),creatine kinase (CK),lactate dehydrogenase (LDH) and glutathione transaminase (AST) in serum were measured by biochemical method.Observation of myocardial histomorphological changes in rats by HE and Mason staining.Results Compared with NC group,the content of FBG,TG,TC,CK,LDH,AST levels in DCM group increased significantly (P<0.05),Heart weight ratio increased (P<0.05),the values of cardiac FS and E/A decreased significantly (P<0.05),LVEF decreased significantly (P<0.05),HE staining could observe the breakage and disorder of myocardial fibers in DCM group,while Masson staining showed the increase of collagen fibers in DCM group.In high dose group,the content of FBG,TG,CK,LDH,AST (P<0.05) decreased,the value of TC and heart weight ratio decreased (P<0.05),cardiac LVEF and FS values increased (P<0.05),and E/A ratio also increased (P<0.05),myocardial injury was lighter than DCM group.Conclusion Berberine can prevent and treat the damage of myocardial structure and function in type 2 diabetic cardiomyopathy rats.
