UPLC-MS法测定藜芦丹参配伍前后吸收转运变化情况

Study of Absorption and Transport of Veratrum Nigrum and Salvia Miltiorrhiza Bunge by UPLC-MS Method

摘要 目的:建立UPLC-MS方法测定藜芦与不同比例丹参配伍前后主要成分经Caco-2细胞吸收转运后的变化情况,从吸收转运的角度分析丹参"叛"藜芦的机制。方法:液质条件为Acquity UPLC BEH C18(1.7μm, 2.1 mm×50 mm)色谱柱,柱温为35℃,流动相为乙腈(A)和0.1%甲酸水溶液(v/v),梯度洗脱;Waters LTQ质谱仪,采用电喷雾电离源,电喷雾电压为5.0 kV,透镜管电压为110 V,金属毛细管电压为40 V、温度为250℃。结果:正离子扫描模式下共鉴定出8种藜芦生物碱,比较藜芦加入不同比例丹参后几种藜芦生物碱峰面积的变化情况及吸收转运的研究,发现丹参是否"叛"藜芦与藜芦的绝对用量和二者的相对用量密切相关。结论:研究结果从化学成分含量变化和吸收转运的角度再次考察了中药"十八反"中丹参"叛"藜芦的配伍禁忌原则,为深入研究中药十八反中藜芦丹参药对奠定基础。 Objective:To establish a UPLC-MS method for the determination of the transportation changes of the main alkaloids from Veratrum nigrum after the combination with Panax ginseng by the Caco-2 monolayer model. The mechanism of Panax ginseng lowering the effect of Veratrum nigrum could be studied from the perspective of absorption and transportation. Methods:The separation was performed on Acquity UPLC BEH C18 column(1.7 mm, 2.1 mm×50 mm) and the column temperature was maintained at 35 ℃. The mobile phase consisted of acetonitrile(A) and 0.1% formic acid aqueous solution(B) with gradient elution. Waters LTQ Mass Spectrometer with ESI ionization source was set with electric spray fog voltage of 5.0 kV, metal capillary voltage of 40 V, lens tube voltage of 110 V, and the metal capillary temperature was 250 ℃. Results: In the positive ion scanning model, 8 kinds of alkaloidsl were identified. And the changes of the peak area and the transportation of several kinds of alkaloids were compared. It showed that whether the Salvia miltiorrhiza Bunge is "rebellious" to Veratrum nigrum is correlated with the relative dosage of the two drugs. Conclusion: In this study, the theory of traditional Chinese medicine was again investigated from the change of chemical content and the transportation and absorption. And the study provided a foundation for the further study of Veratrum nigrum and Salvia miltiorrhiza Bunge.
