Objective The research adopts the method of the Box-Benhnken design combining with response surface analysis to optimize the extraction technology of total flavonoids from Corydalis conspersa. Methods Content of total flavonoids was determined using rutin as the reference and UV spectrometry. Using 60% ethanol as solvent, the influence of ratio of liquid-solid,extraction time and extraction temperature on the extraction yield of flavonoids was evaluated. Results Optimum extraction technology of total flavonoids yield was as follows:ratio of liquid-solid is 500 mL·g-1,extraction temperature is 70 ℃,extracting time is 60 min. Under the optimized extraction conditions,the extraction rate of the total flavonoids was 2.94% and the relative standard odeviation between the predicted(2.99 %)and measured value was 0.27%. Conclusion It is a feasible and stable method to obtain the total flavonoids from the Corydalis conspersa using the ultrasonic optimization extraction technology process combined with Box-Benhnken design and response surface analysis.
