Roux-en-Y胃旁路术对2型糖尿病大鼠模型血清Ghrelin和下丘脑PYY、AgRP表达的影响 预览

Effect of Roux-en-Y gastric bypass on the expression of serum Ghrelin and hypothalamic PYY and AgRP in type 2 diabetic rats

摘要 目的分析Roux-en-Y胃旁路术对2型糖尿病(T2DM)大鼠模型血清胃饥饿素(Ghrelin)和下丘脑肽YY(PYY)、刺鼠相关蛋白(AgRP)表达的影响。方法取24只成年SD雄性大鼠,构建T2DM动物模型,随机分为模型组(未采取手术治疗)与实验组(采取Roux-en-Y胃旁路术),两组各12只,另选取12只健康成年大鼠为对照组。比较三组大鼠术前和术后1周、2周、3周、4周血清总胆固醇(TC)、甘油三酯(TG)、空腹血糖(FBG)及Ghrelin浓度变化,于术后4周时取各组大鼠下丘脑,对PYY和AgRPmRNA相对表达量进行检测。结果术后第2周,实验组血清TC、TG和FBG开始下降,第3周时明显下降,第4周时各指标的水平均基本恢复正常,术后各时间点TC、TG和FBG水平低于模型组,均存在显著差异(P<0.05);模型组术后各时间点TC、TG和FBG水平高于对照组,均存在显著差异(P<0.05)。相比术前,实验组术后第1、2、3、4周Ghrelin水平均下降;相比模型组,实验组且术后第1、2、3、4周Ghrelin水平均显著下降(P<0.01);相比对照组,模型组术后第1、2、3、4周Ghrelin水平均显著升高(P<0.01)。相比对照组,模型组大鼠术后4周下丘脑PYYmRNA相对表达量显著下降,AgRPmRNA相对表达量显著升高(P<0.05);相比模型组,实验组大鼠术后4周下丘脑PYYmRNA相对表达量显著增高,AgRPmRNA相对表达量显著下降(P<0.01)。结论Roux-en-Y胃旁路术可有效调节T2DM大鼠血糖水平,Ghrelin可能通过PYY和AgRP信号调节T2DM能量代谢平衡,进而推测,血清Ghrelin、下丘脑PYY和AgRP信号通路可能具有减轻Roux-en-Y胃旁路术后T2DM病情程度的重要作用。 Objective To analyze the effects of Roux-en-Y gastric bypass on the expression of serum Ghrelin (Ghrelin) and hypothalamic peptide YY (PYY) and agouti-related protein (AgRP) in type 2 diabetes mellitus (T2DM) rats. Methods Twenty-four adult SD male rats were randomly divided into model group (without surgical treatment) and experimental group (with Roux-en-Y gastric bypass). Twelve rats in each group were selected as control group. The changes of serum total cholesterol (TC),triglyceride (TG),fasting blood glucose (FBG) and Ghrelin levels were compared before and after operation in three groups of rats. The hypothalamus of each group was taken at 4 weeks after operation to detect the relative expression of PYY and AgRP mRNA. Results The levels of TC,TG and FBG in the experimental group began to decrease at the 2nd week after operation,and decreased significantly at the 3rd week,and returned to normal at the 4th week ( P <0.05). The levels of TC,TG and FBG at each time point after operation were significantly different from those in the model group ( P <0.05). The levels of TC,TG and FBG at each time point after operation in the model group were significantly different from those in the control group ( P <0.05). Compared with the preoperative group,the Ghrelin level in the experimental group decreased at the 1st,2nd,3rd and 4th week after operation,and decreased significantly at the 1st,2nd,3rd and 4th week after operation ( P <0.01) compared with the model group,and increased significantly at the 1st,2nd,3rd and 4th week after operation compared with the control group ( P <0.01). Compared with the control group,the relative expression of PYY and AgRP in the hypothalamus of rats in the model group decreased significantly 4 weeks after operation,and the relative expression of AgRP increased significantly ( P <0.05). Compared with the model group,the relative expression of PYY and AgRP in the hypothalamus of rats in the experimental group increased significantly 4 weeks after operation,and the relative expression of
