大豆蚜CYP4G51基因克隆及吡虫啉胁迫对其诱导效应 预览

摘要 细胞色素P450是昆虫体内重要的解毒代谢酶,与昆虫对杀虫剂抗性有关。试验通过转录组测序获得1条新的大豆蚜细胞色素P450的cDNA序列,经国际P450命名委员会命名为CYP4G51,该基因GenBank登陆号为MG829857。cDNA 序列全长2 347 bp,含1个长度为1 701 bp开放阅读框,编码566个氨基酸,等电点约为6.46,分子质量约为64.5 ku。氨基酸序列中包含昆虫P450基因共有保守结构域。LC10、LC30和LC50不同浓度吡虫啉诱导3 h后,基因相对表达量均显著上调,与浓度呈正相关;LC50浓度下诱导12 h时相对表达量最高,为未诱导的5.467倍,表明该基因参与大豆蚜对吡虫啉的解毒代谢过程。 Cytochrome P450 was an important detoxification metabolizing enzyme system in insects, it was related to insect resistance to insecticides. In this experiment, one novel cDNA sequence of soybean aphid cytochrome P450 was obtained by transcriptome sequencing, named CYP4G51 by P450 naming committee, which had been registered in GenBank, and the GenBank accession number was MG829857. The full-length cDNA sequence was 2 347 bp, the open reading frame was 1 701 bp and encoded 566 amino acids, the isoelectric point was about 6.46, and the molecular mass was about 64.5 ku. The amino acids sequence included the conserved domain of insect P450. After 3 h of induction with different concentrations of imidacloprid, LC10, LC30 and LC50, the relative expression levels of CYP4G51 was significantly up-regulated and positively correlated with the concentration. The relative expression level was the highest at the induction of LC50 for 12 h treatment, which was 5.467 times with the control. The results indicated that CYP4G51 was involved in the detoxification metabolism of imidacloprid by soybean aphid.
