滋肾育胎丸对大鼠种植窗期子宫内膜容受性的影响及机制 预览

摘要 目的探究滋肾育胎丸对妊娠大鼠子宫内膜容受性的影响和机制。方法对处在胚胎种植窗期的大鼠皮下注射米非司酮溶液,滋肾育胎丸灌胃给药。应用实时荧光定量PCR、Western blot和免疫组化检测子宫组织中LIF、αvβ3的表达以及P38的磷酸化情况,HE染色观察子宫形态,并在D10时计数各组大鼠的胚泡着床数目。结果滋肾育胎丸能够促进胚泡着床。给药后,着床障碍大鼠子宫中LIF和αvβ3的表达量上调,胚泡着床数目增加,同时滋肾育胎丸还能够提高P38的磷酸化水平。结论滋肾育胎丸能够有效改善米非司酮诱导的着床障碍,可能通过活化P38 MAPK通路提高子宫内膜容受性。 Objective:To investigate the effect of Zishen Yutai Pills on endometrial receptivity in pregnant rats and its mechanism.Methods:Pregnant rats were injected with mifepristone solution subcutaneously,followed by intragastric injection of Zishen Yutai Pills.The expression of LIF andαvβ3 and the phosphorylation of P38 in uterine tissues were detected by Real-time PCR,Western blot and Immunohistochemistry.HE staining was used to detect the uterine morphology.The numbers of blastocysts were counted at D10.Result:Zishen Yutai Pills could promote blastocyst implantation.After injection,the expression numbers of LIF andαvβ3 were up-regulated in rats with embryo implantation dysfunction and the number of blastocyst implantation was increased significantly.Zishen Yutai Pills also improved the phosphorylation of P38.Conclusion:Zishen Yutai Pills can effectively improve mifepristone-induced embryo implantation dysfunction and may improve endometrial receptivity by activating P38 MAPK pathway.
