骨膜源性细胞和骨髓间充质干细胞直接共培养的实验研究 预览

摘要 目的探讨骨膜源性细胞(PDCs)和骨髓间充质干细胞(BMSCs)不同比例下直接共培养对细胞的增殖及成骨分化的影响,优选细胞直接共培养的适宜比例。方法取PDCs和BMSCs,在保证每组总细胞数一致的情况下,分别以PDCs和BMSCs按2∶1、1∶1、1∶2、1∶0、0∶1比例混合培养,通过细胞增殖能力,碱性磷酸酶活性(alkaline phosphatase activity, ALP)和矿化结节形成的测定,来观察PDCs和BMSCs不同比例共培养下细胞增殖和成骨分化能力。结果 PDCs和BMSCs直接共培养(2∶1、1∶1、1∶2)的细胞增殖能力高于单独PDCs组(1∶0组)和单独BMSCs组(0∶1组),但差异无统计学意义(P>0.05),共培养组细胞ALP表达较对照组高(P<0.05),2∶1组ALP表达最高(P<0.05),同时共培养组矿化结节形成较多,1∶0组和0∶1组矿化结节形成较疏散。结论 PDCs和BMSCs直接共培养有利于细胞的增殖能力和成骨能力,以2∶1比例直接共培养时最佳,适宜进一步的组织工程研究。 Objective To investigate the effects of direct co-culture ratio on the proliferation and osteogenic differentiation of PDCs and BMSCs, and to optimize the co-culture ratio.Methods PDCs and BMSCs were cultured at the ratio of 2∶1, 1∶1,1∶2,1∶0,0∶1 respectively, and the total number of cells in each group was the same.The proliferation and osteogenic differentiation of PDCs and BMSCs co-cultured were observed by cell proliferation, alkaline phosphatase activity(ALP), and mineralized nodules formation.Results The cell proliferation of the co-culture group were higher than that of 1∶0 and 0∶1 groups, but there was no statistical differences between all groups(P>0.05).The ALP expression level of the co-culture group(2∶1,1∶1 and 1∶2 ratio) especially in 2∶1 ratio expressed higher than that of control groups(1∶0 and 0∶1 ratio)(P<0.05), and the mineralized nodules in co-culture groups were formed more than that in the control groups(1∶0 and 0∶1 ratio).Conclusion When PDCs and BMSCs was directly co-cultured at 2∶1 ratio, the total cell proliferation and osteogenic ability are stronger than other groups, which are suitable for further tissue engineering research.
