NCOA2促进急性B淋巴细胞白血病细胞增殖及下游信号通路的生物信息学分析

NCOA2 promoting the proliferation of acute B lymphoblastic leukemia cells and bioinformatics analysis of the downstream signaling pathway

摘要 目的探讨核受体辅激活因子2(Nuclear Receptor Coactivator2,NCOA2)在急性B淋巴细胞白血病(B-ALL)细胞增殖中的作用,并利用生物信息学方法探究NCOA2促进B-ALL细胞增殖涉及的基因和信号通路。方法在B-ALL细胞Nalm6及BALL-1中使用siRNA技术敲减NCOA2,通过Western blot检测NCOA2的敲除效率,通过EdU方法检测Nalm6及BALL-1细胞的增殖能力变化;使用GEO2R软件分析NCOA2敲减细胞系的表达谱数据(GSE118992),找到有意义的差异基因;使用DAVID和KOBAS软件对差异基因进行GO和KEGG分析,对差异基因的作用和参与的信号通路进行聚类分析;使用Venny2.1软件对GO和KEGG中参与调控细胞增殖的因子进行并集分析;使用STRING软件分析参与NCOA2调节细胞增殖相关基因之间的相互作用关系。结果NCOA2siRNA能够抑制NCOA2蛋白表达和Nalm6及BALL-1细胞的增殖能力,同时筛选出受其调控的差异显著性基因322个,选择排在前十位的生物学功能和信号通路,把其中参与调节细胞增殖的基因取并集,找到受NCOA2调控的参与细胞增殖的4个基因(TGFBR2、CRKL、CCND2、PDGFRA),发现它们之间具有相关作用关系。结论NCOA2调节细胞增殖相关因子(TGFBR2、CRKL、CCND2、PDGFRA),参与B淋巴细胞白血病发生发展。 Objective To study the effect of NCOA2 in the cell proliferation of acute B lymphoblastic leukemia and to analyze the relationship of genes with related signaling pathways by bioinformatics methods. Methods NCOA2 was knocked down by siRNA in Nalm6 cells and BALL-1 cells, and the knockout efficiency of NCOA2 was detected by Western blot, and the proliferation ability of Nalm6 cells and BALL-1 cell was detected by EdU method. The Expression profile data(GSE118992) in NCOA2 knockdown cell line was analyzed by GEO2 R software, DAVID and KOBAS software were used to perform GO and KEGG analysis on the differential genes, Venny2.1 software was used to find out the same factors involved in the regulation of cell proliferation in KEGG, and GO. STRING software was used to analyze the interaction between the genes. Results NCOA2 siRNA inhibited the expression of NCOA2 and the proliferation of Nalm6 and BALL1 cells, 322 genes were screened out in the NCOA2 knockdown expression profiling, the top ten biological functions and signaling pathways were selected, 4 genes(TGFBR2、 CRKL、 CCND2、 PDGFRA) were found in the role of regulating the cell proliferation and regulated by NCOA2, and have the related relationships among them. Conclusion NCOA2 plays a role of promoting cell proliferation by regulating the expression of cell proliferation related factors(TGFBR2、CRKL、CCND2、PDGFRA) and involved in the pathogenesis and malignant progression of acute B lymphoblastic leukemia.
