重铬酸钾染毒对发育期大鼠学习记忆及海马氧化应激水平的影响 预览

Effect of Potassium Dichromate on Learning-Memory Ability and Hippocampal Oxidative Stress in Developing Rats

摘要 目的观察重铬酸钾对大鼠学习记忆能力和海马组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的影响。方法健康雄性SD大鼠40只,随机分为四组:对照组(蒸馏水),重铬酸钾低浓度(0. 8 mg/kg)组、重铬酸钾中浓度(4. 0 mg/kg)组和重铬酸钾高浓度(20. 0 mg/kg)组,每组10只,持续染毒6个月。Morris水迷宫实验检测大鼠的学习记忆能力,分光光度法测定大鼠海马组织SOD活性和MDA含量。结果各组大鼠的平均回避潜伏期不同,差异具有统计学意义(F=28. 781,P=0. 000),重铬酸钾低、中、高浓度组大鼠的平均躲藏潜伏期长于对照组(P<0. 05),重铬酸钾各组之间的差异均具有统计学意义(P<0. 05);各组大鼠穿越平台次数及原平台象限的停留时间的差异均具有统计学意义(F=29. 959,P=0. 000;F=32. 649,P=0. 000),且各组间的两两比较差异均具有统计学意义(P<0. 05),随给药浓度的增加而减少。各组大鼠海马组织中的SOD活性的差异具有统计学意义(F=43. 051,P=0. 000),重铬酸钾低、中、高浓度组大鼠的SOD活性均低于对照组(P<0. 05);各组大鼠海马组织中的MDA含量的差异具有统计学意义(F=397. 261,P=0. 000),重铬酸钾低、中、高浓度组大鼠的MDA含量均高于对照组(P<0. 05)。结论重铬酸钾能够使大鼠的学习记忆能力降低,SOD活性下降,MDA含量升高,其机制可能与重铬酸钾中六价铬诱导的氧化应激有关。 Objective To observe the effect of potassium dichromate on the learning-memory ability,the activity of superoxide dismutase( SOD),and the content of malondialdehyde( MDA) in hippocampus in rats. Methods 40 healthy male SD rats were randomly divided into 4 groups,a control group( distilled water),a group of low concentration of potassium dichromate( 0. 8 mg/kg),a group of medium concentration of potassium dichromate( 4 mg/kg),and a group of high concentration of potassium dichromate( 20 mg/kg),with 10 rats in each group. The latter 3 groups were treated with intragastric administration of potassium dichromate for 6 months. Morris water maze test was used to detect the learning-memory ability of the rats. SOD activity and MDA content in hippocampus of the rats were determined by spectrophotometry. Results The average escape latency of the rats varied in different groups,with statistically significant difference( F = 28. 781,P = 0. 000). The average escape latency of the rats in the group of low,medium and high concentration potassium dichromate was longer than that in the control group( P<0. 05),and the difference among the 3 groups of potassium dichromate was statistically significant( P<0. 05). The difference in the times of crossing platform and the residence time of the original platform quadrant among the 4 groups was statistically significant( F = 29. 959,P = 0. 000,F = 32. 649,P = 0. 000),and so was the difference of pairwise comparison among the groups( P<0. 05). Both of the times of crossing platform and the residence time of the original platform quadrant decreased with the increase of drug concentration. The difference in SOD activity in the hippocampus of the rats among the 4 groups remained statistically significant( F = 43. 051,P = 0. 000). The activity of SOD in the groups of low,medium and high concentration of potassium dichromate was lower than that of the control group( P < 0. 05).
