麦粒灸对双转基因AD小鼠GFAP与Aβ1-42斑块共定位的影响 预览

Effect of Wheat-sized Moxibustion on the Co-localization of GFAP and Aβ1-42 Plaque in Double-transgenic AD Mice

摘要 目的探讨麦粒灸治疗阿尔茨海默病(AD)的相关机制及对小鼠脑内胶质纤维酸性蛋白(GFAP)与b-淀粉样蛋白1-42(Aβ1-42)斑块的共定位的影响。方法 PCR法进行小鼠基因型的鉴定,将1.5月龄双转基因AD小鼠40只,随机分为治疗组和模型组各20只,同龄、同背景的阴性纯合子小鼠20只,为正常组。治疗组小鼠取双侧心俞、肾俞行麦粒灸治疗。采用免疫荧光法检测AD转基因小鼠GFAP与Aβ1-42斑块的共定位情况。结果模型组AD小鼠额叶皮层和海马区GFAP与Aβ1-42阳性细胞共表达率明显高于正常组,差异有统计学意义(P<0.05);治疗组AD小鼠治疗后额叶皮层、海马区GFAP与Aβ1-42阳性细胞共表达率低于模型组,差异有统计学意义(P<0.05)。结论早期麦粒灸疗法可以抑制AD小鼠星形胶质细胞的炎症反应,抑制Ab异常聚集,延缓AD病理过程的进展。 Objective To discuss the mechanism of wheat-sized moxibustion in treatment of Alzheimer’s disease(AD)and its effect on the co-localization of glial fibrillary acidic protein(GFAP)and b-amyloid protein1-42(Aβ1-42)plaque in rat models.Method The genotypes of the mice were identified by polymerase chain reaction(PCR).Forty 1.5-month-old double-transgenic AD mice were randomized into a treatment group and a model group,with 20 mice in each group.Twenty negative homozygous mice at the same age and with the same background were taken as the normal group.Mice in the treatment group were treated with wheat-sized moxibustion at bilateral Xinshu(BL15)and Shenshu(BL23).Immunofluorescence was used to detect the co-localization of GFAP and Aβ1-42 plaque in the transgenic AD mice.Result The co-expression rate of GFAP and Aβ1-42 positive cells in the frontal cortex and hippocampus of AD mice in the model group was significantly higher than that in the normal group(P<0.05);the co-expression rate of GFAP and Aβ1-42 positive cells in the frontal cortex and hippocampus of AD mice in the treatment group was significantly lower than that in the model group(P<0.05).Conclusion Early-stage intervention with wheat-sized moxibustion can inhibit the inflammatory response mediated by astrocytes in AD mice,restrain the abnormal accumulation of b-amyloid protein and slowdown the development of AD.
