紫草素诱导人乳腺癌MCF-7细胞凋亡及其机制探讨 预览

摘要 目的探讨紫草素对人乳腺癌MCF-7细胞凋亡的影响及其机制。方法采用不同浓度(0.1、1、10μmol/L)的紫草素处理人乳腺癌MCF-7细胞24h,观察细胞活性、凋亡细胞比例及凋亡相关蛋白表达的变化。结果紫草素能明显抑制MCF-7细胞增殖,促进细胞凋亡,升高Caspase-3、Cleaved Caspase-3、Caspase-9和Cleaved Caspase-9表达水平。结论紫草素通过促进凋亡相关蛋白表达诱导MCF-7细胞凋亡,有望成为治疗乳腺癌的有效药物。 Objective To investigate the effect and mechanism of shikonin on human breast cancer MCF-7 cells apoptosis and its mechanism.Method Human breast cancer MCF-7 cells were treated with different concentrations of shikonin(0.1,1,10μmol/L)for 24h.Cell activity,apoptosis cell ratio and expressions of apoptosis protein were detected.Result Shikonin could obviously inhibit MCF-7 cells’proliferation and promote apoptosis,increased the expression of Caspase-3,Cleaved Caspase-3,Caspase-9 and Cleaved Caspase-9.Conclusion Shikonin induced apoptosis of MCF-7 cells by promoting expression of apoptosis related proteins,shikonin is expected to be a useful drug for the treatment of laryngeal cancer.
