黄芩苷对慢性心肌衰竭大鼠心肌细胞凋亡及相关信号通路表达的影响

Effect of Baicalin on myocardial cell apoptosis and Akt/AMPK/mTOR signal pathway expression in chron⁃ic myocardial failure rats

摘要 目的慢性心力衰竭仍是未能攻克的课题,中药治疗是我们探讨的重要方向。文章旨在研究黄芩苷对慢性心肌衰竭大鼠心肌细胞凋亡及Akt/AMPK/mTOR信号通路表达影响。方法SPF级雄性SD大鼠60只,随机分为正常组、模型组、阳性对照组和实验组,每组各15只。阳性对照组给予氯沙坦40 mg/kg灌胃,1次/d;实验组给予黄芩苷50 mg/kg灌胃,1次/d;正常组和模型组灌胃等剂量等渗盐水,1次/d。各组连续灌胃4周。HE染色观察心肌细胞病理形态学;心肌组织细胞指数(CAI)以原位末端标记法检测;心肌Bax、Bcl⁃2蛋白表达以免疫组化法检测;心功能以心脏超声检测;Akt、AMPK、mTOR mRNA表达以RT⁃PCR反应测定。结果与正常组CAI(1.27±0.21)比较,模型组、阳性对照组和实验组(65.38±7.89、23.69±7.53、25.05±8.91)升高(P<0.05);而阳性对照组和实验组较模型组降低(P<0.05)。与正常组Bcl⁃2蛋白表达(149.83±14.32)比较,模型组、阳性对照组和实验组(102.64±8.98、133.73±12.10、135.62±10.87)下降(P<0.05);而阳性对照组和实验组较模型组升高(P<0.05)。与正常组Bax蛋白表达(134.25±18.97)比较,模型组、阳性对照组和实验组(218.73±15.42、163.29±23.15、162.10±27.36)增加(P<0.05);阳性对照组和实验组Bax蛋白表达较模型组下降(P<0.05)。与正常组LVEF(50.83±2.14)比较,模型组、阳性对照组和实验组(34.25±2.87、39.25±2.01、42.34±1.90)明显下降P<0.05);阳性对照组和实验组LVEF较模型组升高(P<0.05)。模型组、阳性对照组和实验组(1.39±0.16、1.15±0.14、0.93±0.10)较正常组LVD(0.74±0.13)增加(P<0.05);阳性对照组和实验组LVD和LVS较模型组下降(P<0.05)。模型组、阳性对照组和实验组(0.96±0.12、0.78±0.10、0.69±0.12)正常组LVS(0.57±0.13)增加(P<0.05);阳性对照组和实验组LVS较模型组下降(P<0.05)。与正常组比较,模型组、阳性对照组和实验组Akt、AMPK、mTOR mRNA表达降低(P<0.05);阳性对照组� Objective To investigate the effect of Baicalin on the apoptosis of cardiomyocytes and the expression of Akt/Amp activated protein kinase(AMPK)/mammalian rapamycin target protein(mTOR)signaling pathway in rats with chronic myocardial fail⁃ure.Methods Sixty male SD rats of SPF grade were randomly divided into normal group,model group,positive control group and experimental group,with 15 rats in each group.Rats in the positive control group were given 40mg/kg losartan once a day.Rats in the ex⁃perimental group were given 50mg/kg Baicalin once a day.Rats in the normal group and the model group were given the same dose of saline once a day.Each group was given gavage for 4 weeks.HE staining was used to observe the pathomorphology of cardiomyocytes.The myo⁃cardial tissue index(CAI)was detected by in situ end labeling.The expressions of Bax and bcl⁃2 protein in myocardium were detected by immunohistochemistry.The cardiac function was detected by echocardiography.The expression of Akt,AMPK and mTOR mRNA was detected by RT⁃PCR.Results Compared with the normal group,the apoptosis indexes of the model group,the positive control group and the experimental group were increased(P<0.05).Compared with the model group,the apoptosis indexes of the positive con⁃trol group and the experimental group were decreased(P<0.05),while there was no significant difference between the positive control group and the experimental group(P>0.05).Compared with the model group,the expression of Bcl⁃2 protein was increased and Bax protein was decreased in the positive control group and the experimental group(P<0.05),while there was no significant difference be⁃tween the positive control group and the experimental group(P>0.05).Compared with the normal group,the LVEFs of the model group,the positive control group and the experimental group were decreased,while the LVD and LVS were increased(P<0.05).Com⁃pared with the model group,the LVEFs of the positive control group and the experimental group were increased,while the LVD and LVS
