摘要
目的 筛选四妙勇安汤具有过氧化物酶体增殖物激活受体γ(PPAR-γ)激活作用的活性部位,并对其化学成分进行分析,探讨提取物的活性成分和抗炎作用.方法 采用醇水提取、树脂分离获得四妙勇安汤的8个提取物,以PPAR-γ为靶点,于细胞水平筛选四妙勇安汤提取物的活性部位,对有活性趋势的提取物绘制剂量依赖性曲线,计算EC50值以及相对活性,并采用液相色谱-串联质谱法对活性部位的化学成分进行分析鉴定.结果 PPAR-γ激动模型中,提取物CS04及CS05具有较强的PPAR-γ激活作用.对活性部位CS04和CS05进行化学成分鉴定,共鉴别出31个成分.结论 四妙勇安汤提取物CS04和CS05具有较强激活PPAR-γ作用,这一作用可能与其抑制动脉粥样硬化炎症有关.
Objective To screen the active parts of Simiao Yong′an Decoction with peroxisome proliferators-activated receptorγ(PPAR-γ)activation and analyze its chemical components,and to explore the active components and antiinflammatory effects of the extract.Methods Eight alcohol extracts of Simiao Yong′an Decoction were obtained by alcohol extraction and resin separation.The PPAR-γwas used as the target to screen the active parts of the extract of Simiao Yong′an Decoction at the cell level.A dose-dependent curve was drawn for the extracts with active tendency.EC50 value and relative activity value were calculated,and the chemical composition of active parts was analyzed and identified by liquid chromatograph-tandem mass spectrometer technology.Results In the PPAR-γactivation model,the extracts CS04 and CS05 have strong PPAR-γactivation.The chemical components of the active parts CS04 and CS05 with strong PPAR-γagonistic effects were identified.A total of 31 components were identified.Conclusion CS04 and CS05 of Simiao Yong′an Decoction have strong PPAR-γactivation,which may be related to the inhibition of atherosclerosis inflammation.
