mTOR-S6K1-Gli1信号通路在小鼠慢性瘙痒中的作用

摘要 目的:探讨哺乳动物雷帕霉素靶蛋白-核糖体蛋白S6激酶1-胶质瘤相关癌基因同源蛋白-1(mTOR-S6K1-Gli1)信号通路在慢性瘙痒中的作用。方法:取雄性昆明小鼠40只,分为空白对照组、溶剂对照组、致痒组、致痒+抑制剂组和致痒+抑制剂溶剂组,每组8只。后3组小鼠采用定期反复予恶唑酮溶液(乙醇为溶剂)涂抹腰背部皮肤的方法建立慢性瘙痒模型,溶剂对照组给予乙醇,致痒+抑制剂组和致痒+抑制剂溶剂组除致痒处理外,分别给予GANT61(Gli1抑制剂)溶液和相应溶剂鞘内注射。实验第0、7、9、12、14、16、19天记录涂抹恶唑酮溶液后30 min内小鼠搔抓次数;实验第19天,5组小鼠各取3只处死,取给药处皮肤行HE染色,观察局部病理变化,同时用Western blot法测定脊髓组织Gli1蛋白的表达;取溶剂对照组和致痒组小鼠各3只,采用免疫组化法观察mTOR、S6K1、Gli1蛋白在脊髓组织的定位。结果:与空白对照组和溶剂对照组相比,致痒组小鼠搔抓次数增加(P<0.05),局部皮肤出现广泛炎症细胞浸润,提示建模成功。与致痒组相比,致痒+抑制剂组小鼠搔抓次数减少(P<0.05),皮肤组织炎症细胞减少。与溶剂对照组相比,致痒组脊髓组织Gli1蛋白的表达升高(P<0.05),而致痒+抑制剂组无明显变化(P>0.05)。免疫组化结果显示mTOR、S6K1、Gli1主要表达在致痒组小鼠脊髓背角浅层。结论:mTOR-S6K1-Gli1信号通路可能参与了慢性瘙痒的发生。 Aim:To investigate the role of the mTOR-S6K1-Gli1 signaling pathway in chronic pruritus.Methods:Forty male Kunming mice were allocated into 5 groups,blank control group,solvent control group,itching group,itching+inhibitor group and itching+inhibitor solvent group(8 mice in each group).The chronic pruritus model was made by periodically applying oxazolone solution(ethanol as solvent)on the waist skin.The mice in the latter 3 groups were periodically given oxazolone solution,and the scratching times during 30 min on the induction day,7th,9th,12th,14th,16th,and 19th day were recorded.On the 19th day,3 mice were taken,the skin tissue was prepared for HE staining to observe the histopathological changes;the spinal cord tissue was taken,and the expression of Gli1 was detected by Western blot.Then 3 mice in the solvent control group and itching group were taken,and the location of mTOR,S6K1 and Gli1 was detected by immunohistochemistry.Results:Compared with blank control group and solvent control group,the scratching times increased(P<0.05)and extensive inflammatory cell infiltration of localized skin was observed in itching group.Compared with itching group,the scratching times decreased(P<0.05)in itching+inhibitor group,and the inflammatory cell infiltration alleviated.Compared with solvent control group,the expression of Gli1 in itching group was increased(P<0.05)while that in the itching+inhibitor group had no significant changes(P>0.05).The above 3 proteins mainly expressed in the superficial layer of the dorsal horn of the spinal cord of the mice in the itching group.Conclusion:The mTOR-S6K1-Gli1 signaling pathway may be involved in chronic pruritus.
