非受体酪氨酸激酶BMX对宫颈癌C-33 A细胞成球能力的影响

摘要 为探索非受体酪氨酸激酶BMX(也称Etk)对宫颈癌C-33 A细胞成球能力的影响,应用免疫细胞化学和Western blot检测BMX在宫颈癌细胞系HeLa、SiHa、C-33 A、HT-3、CaSki中的表达及定位,构建BMX过表达质粒载体,转染至C-33 A细胞中,筛选稳定过表达的细胞株,利用细胞成球实验检测BMX对C-33 A细胞成球能力的影响。免疫细胞化学染色显示,BMX主要在细胞浆中表达,在细胞核中也有部分表达,在HeLa、SiHa、HT-3及CaSki细胞系中BMX高表达,而在C-33A细胞中低表达。Western blot检测也得出同样的结果。将BMX过表达质粒载体转染至C-33A细胞系中,鉴定稳定过表达的细胞株(C-33 A-AcGFP,C-33 A-BMX)。对照组和BMX过表达组细胞成球实验显示,对照组球体较为松散而过表达组较为密实,两组细胞成球率无明显差异(P>0.05),但是过表达组球体直径明显大于对照组(P<0.001)。上述结果表明:非受体酪氨酸激酶BMX促进宫颈癌C-33 A细胞的成球能力。 This paper is to explore the effects of non-receptor tyrosine kinase BMX on the pelletization ability of C-33 A cells in cervical cancer.Immunocytochemistry and Western blot were used to test the expression and positioning of BMX in cervical cancer cell lines HeLa,SiHa,C-33 A,HT-3,and CaSki.BMX hyperexpression plasmid vector was built to transfect to C-33 A cells for screening stable cell lines of hyperexpression.Cell pelletization experiment was used to detect the effect of BMX on the pelletization ability of C-33 A cells.Immunocytote chemical staining shows that BMX is expressed mainly in cell plasma and partly in cell nucleus,at high levels in HeLa,SiHa,HT-3 and CaSki cell lines and at a low level in C-33 A cells.The same results were obtained by Western blot.The BMX hyperexpression plasmid vector was transfected to C-33 A cell line to identify stable cell strains of hyperexpression(C-33 A-AcGFP,C-33 A-BMX).The pelletization test of control group and BMX hyperexpression group showed that the sphere of control group was looser than that of the hyperexpression group,and rate of pelletization of two groups have no significant difference(P>0.05),but the diameter of sphere of the hyperexpression group is much greater than that of the control group(P<0.001).These results show that non-receptor tyrosine kinase BMX promotes the pelletization capacity of C-33 A cells in cervical cancer.
