在水温19~23℃下,将平均体质量为(10±1)g的泥鳅Misgurnus anguillicaudatus分别放在冰片浓度7g/L、8g/L、9g/L、10g/L、11g/L、12g/L、13g/L、14g/L、15g/L、16g/L和17g/L的2L容器中,每组3尾鱼,3个平行。每组泥鳅深度麻醉10min后,尾静脉采血。每组血量达25mL,放入4℃冰箱中冷藏4h,1000r/min离心10 min至完全分层,取上清液,测定血红蛋白含量和过氧化氢酶活力,研究了冰片的麻醉效果及其对血液免疫指标的影响。结果显示,随着冰片浓度的增加,泥鳅进入麻醉的时间缩短,呼吸频率逐渐减缓,摆尾次数逐渐减少,复苏时间越长。当冰片浓度达到15g/L以上时,泥鳅应激反应剧烈,开始死亡。冰片浓度继续加大,泥鳅迅速进入深度麻醉。当冰片浓度达到17g/L时,泥鳅全部死亡;泥鳅血液中的血红蛋白含量变化不明显,过氧化氢酶的活性随冰片浓度的升高而降低。
Loach(Misgurnus anguillicaudatus)with an average body weight of(10±1)g were placed in 2 L basins with borneol concentrations of 7 g/L, 8 g/L, 9 g/L, 10 g/L, 11 g/L, 12 g/L, 13 g/L, 14 g/L, 15 g/L, 16 g/L and 17 g/L at a density of 3 fish per basin with triplication at water temperature of 19 ~ 23℃, and 25 mL blood was sampled from caudal veins of the loach with 10 minutes of deep anesthesia in each group, refrigerated for 4 hours at 4 ℃, centrifuged for 10 minutes at 1000 r/min until completely stratified. The supernatant was taken and the hemoglobin content and catalase activity were determined to investigate the anesthetic effect of borneol and influence of borneol on blood immune indices. It was found that it took shorter time for the loach to enter the anesthetic, and the longer time for the recovery, with lower respiratory rate and less tail beating frequency, with the increase in concentration of the borneol. The loach showed violent response to borneol stress, even began to die at borneol concentration of 15 g/L or more. As the concentration of borneol was increased to 17 g/L, the loach immediately turned into deep anesthesia, and all died. There was no significant difference in blood hemoglobin content in the loach exposed to various borneol concentrations, even though catalase activity was decreased slightly with the increase in the anesthetic concentration.
