丹蒌片对Aβ1-40致脑微血管内皮细胞损伤模型活性的影响 预览

摘要 目的:观察丹蒌片对β淀粉样蛋白(β-Amyloid protein,Aβ)致脑微血管内皮细胞(brain microvascular endothelial cells,BMEC)损伤模型活性的影响。方法:培养BMEC,用Aβ1-40造成BMEC损伤,加入不同剂量的丹蒌片含药血清,6 h、12 h、24 h后用CCK8染色法检测细胞活性,并检测细胞培养液中乳酸脱氢酶(lactic dehydrogenase,LDH)的漏出量。结果:与正常对照组比较,模型组细胞活性降低,细胞培养上清液中LDH释放量增加(P<0.01)。与模型组比较,12 h后各剂量丹蒌片含药血清组细胞活性显著升高,12 h细胞活性高于24 h的细胞活性(P<0.05);丹蒌片含药血清各组细胞培养上清液中LDH水平降低,12 h上清液中LDH水平显著低于6 h、24 h(P<0.05);高剂量、中剂量含药血清组细胞上清液LDH水平显著降低(P<0.05)。结论:丹蒌片能提高Aβ1-40致BMEC损伤后细胞的活性,减少细胞LDH的漏出量,起到保护脑微血管内皮的作用。 Objective:To observe the effect of Danlou tablet on the activity of brain microvascular endothelial cells(BMEC)injury model induced byβ-Amyloid protein(Aβ).Methods:BMEC was cultured and damaged by Aβ1-40.Different doses of drug-containing serum of Danlou tablets were added.Cell viability was detected by CCK8 staining after 6,12 and 24 hours.The leakage of lactic dehydrogenase(LDH)in cell culture medium was also detected.Results:Compared with the normal control group,the cell activity of the model group decreased and the LDH release in the supernatant of cell culture increased(P<0.01).Compared with the model group,the cell activity of the drug-containing serum of Danlou tablets increased significantly after 12 hours,and the cell activity at 12 hours was higher than that at 24 hours(P<0.05);the LDH level in the cell culture supernatant of the drug-containing serum of Danlou tablets decreased,and the LDH level in the supernatant at 12 hours was significantly lower than that at 6 hours and 24 hours(P<0.05);the high and medium dose of the drug-containing serum of Danlou tablets were significantly lower than that at 6 hours and 24 hours(P<0.05).Conclusion:Danlou tablets can improve the activity of BMEC cells after Aβ1-40 induced injury,reduce the leakage of LDH,and protect brain microvascular endothelium.
